枸杞多糖抑制Fas/FasL途径减轻高糖诱导的足细胞炎症损伤及凋亡的机制研究

摘  要:探究枸杞多糖通过调控脂肪酸合酶(Fas)/脂肪酸合酶配体(FasL)信号通路对D-葡萄糖诱导的人肾小球足细胞炎症和凋亡的影响。体外培养人肾小球足细胞HGPC细胞系,将实验分为预实验和正式实验。预实验分为对照组、高糖组(30 mmol/L D-葡萄糖)、低/中/高浓度组;正式实验分为对照组、高糖组、枸杞多糖组、ZB4组、抑制剂组、激活剂组,干预24 h。检测细胞活力、凋亡率、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)及Fas/FasL通路相关蛋白的表达水平。预实验中,高糖组与对照组相比,细胞IL-6、TNF-α和Caspase-3蛋白水平升高(P<0.05),细胞活力降低(P<0.05);高浓度组与高糖组相比,细胞IL-6、TNF-α和Caspase-3蛋白水平降低(P<0.05),而细胞活力升高(P<0.05)。所以确定150μg/mL枸杞多糖为最佳作用浓度,进行后续正式实验。正式实验中,高糖组与对照组相比,细胞IL-6、TNF-α、凋亡率、Caspase-3、Fas和FasL水平升高(P<0.05);枸杞多糖组和ZB4组与高糖组相比,细胞IL-6、TNF-α、凋亡率、Caspase-3、Fas和FasL水平降低(P<0.05);在枸杞多糖的基础上加Fas/FasL通路抑制剂ZB4后,细胞IL-6、TNF-α、凋亡率、Caspase-3、Fas和FasL表达水平进一步降低(P<0.05),加Fas/FasL通路激活剂CH11后,显著扭转了上述指标的变化趋势(P<0.05)。枸杞多糖可通过阻断Fas/FasL通路的信号转导抑制D-葡萄糖诱导的人肾小球足细胞HGPC细胞炎症反应和凋亡。To explore the effects of lycium barbarum polysaccharide(LBP)on D-glucose-induced inflammation and apoptosis of human glomerular podiocytes by regulating fatty acid synthase(Fas)/fatty acid synthase ligand(FasL)signaling pathway,human glomerular podocyte HGPC cell line was cultured in vitro.The experiment was divided into pre-experiment and formal experiment.Pre-experiment was divided into control group,high glucose group(30 mmol/L D-glucose),low/medium/high concentration group.The formal experiment was divided into control group,high glucose group,lycium barbarum polysaccharide group,ZB4 group,inhibitor group,activator group,intervention for 24 h.Cell viability,apoptosis rate,expression levels of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),cysteine aspartate protease-3(Caspase-3)and Fas/FasL pathway related protein were measured.In the pre-experiment,compared with the control group,the levels of IL-6,TNF-αand Caspase-3 protein were increased(P<0.05),and the cell viability was decreased(P<0.05).Compared with high glucose group,the levels of IL-6,TNF-αand Caspase-3 protein in high concentration group were decreased(P<0.05),while cell viability was increased(P<0.05).Therefore,150μg/mL of lycium barbarum polysaccharide was determined as the optimal concentration,and subsequent formal experiments were conducted.In the formal experiment,the levels of IL-6,TNF-α,apoptosis rate,Caspase-3,Fas and FasL were increased in the high glucose group compared with the control group(P<0.05).The levels of IL-6,TNF-α,apoptosis rate,Caspase-3,Fas and FasL were decreased in lycium barbarum polysaccharide group and ZB4 group compared with high glucose group(P<0.05).After addition of Fas/FasL pathway inhibitor ZB4 on the basis of lycium barbarum polysaccharide,the expression levels of IL-6,TNF-α,apoptosis rate,Caspase-3,Fas and FasL were further decreased(P<0.05).After addition of Fas/FasL pathway activator CH11,the change trend of the above indexes was significantly reversed(P<0.05).Lycium barbarum polysaccharide could i
