摘 要:目的:优选毛桃血藤颗粒的制备工艺,建立相关薄层色谱(TLC)鉴别方法。方法:以补骨脂素含量和得膏率的综合评分为评价指标,采用单因素试验结合Box-Behnken响应面法考察提取工艺;以颗粒制粒、成型情况为评价指标考察成型工艺。采用TLC法对颗粒中的五指毛桃、覆盆子进行定性鉴别。结果:最佳提取工艺为加水量12倍,浸泡50 min,提取时间70 min,提取2次;最优成型工艺为药辅比1∶1.5,辅料为可溶性淀粉∶甘露醇(1∶1),85%乙醇润湿,制粒,70℃烘干。TLC斑点清晰,阴性无干扰。结论:研究的制备工艺和薄层鉴别方法稳定、可行,可为后期开发生产提供实验基础。Objective To optimize the preparation process of Maotao Xueteng granules and to establish the related thin-layer chromatography(TLC)identification method.Methods The extraction process was investigated by single-factor test combined with Box-Behnken response surface method using the combined scores of the content of psoralen and the rate of obtaining paste as evaluation indexes;The pellet granulation and molding situation was used as an evaluation index to examine the molding process.TLC method to identify Ficus hirta Vahland Rubus chingii Huin the granules.Results The optimum preparation process was as follows:add 12 times of water and soak for 50 min,and decoction twice with one hour for each time;The optimal molding process was to add mixed excipients of soluble starch∶mannitol(1∶1)according to the drug-excipient ratio of 1∶1.5,and add 85%ethanol for granulation,drying at 70℃.TLC results are well spotted and negative without interference.Conclusion The preparation process and thin-layer identification method studied are stable and feasible,which can provide an experimental basis for later development and production.
