摘  要:目的探讨脑蛋白水解物-Ⅰ(CH-Ⅰ)对D-半乳糖(D-Gal)致PC12细胞衰老的影响及其机制。方法将PC12细胞分为对照组(常规培养)、模型组(D-Gal诱导)、低浓度组和高浓度组(在模型组基础上分别给予60、120 mg/L的CH-Ⅰ)。利用β-半乳糖苷酶(SA-β-Gal)染色法检测各组PC12细胞衰老率,采用流式细胞术检测各组细胞的凋亡率,采用Western Blot方法检测各组细胞中p-STAT3、TERT相对表达水平,采用PCR-ELISA法检测各组细胞中端粒酶的活性。结果与对照组相比,模型组细胞衰老率和凋亡率显著升高(t_(LSD)=27.03、17.77,P<0.05),端粒酶活性显著下降(t_(LSD)=6.21,P<0.05)。与模型组相比,低、高浓度组SA-β-Gal细胞衰老率和凋亡率均明显降低(t_(LSD)=7.74~21.43,P<0.05),端粒酶活性均显著升高(t_(LSD)=2.01、2.49,P<0.05),细胞中TERT、p-STAT3相对表达水平显著升高(t_(LSD)=2.21~6.03,P<0.05)。结论CH-Ⅰ可能是通过促进STAT3磷酸化,上调PC12细胞的TERT表达和端粒酶活性,从而减轻D-Gal诱导的PC12细胞衰老。Objective To investigate the effect and mechanism of cerebroprotein hydrolysate-Ⅰ(CH-Ⅰ)on senescence of PC12 cells induced by D-galactose(D-Gal).Methods PC12 cells were divided into control group(routine culture),model group(induced by D-Gal),low concentration group(treated with 60 mg/L CH-I in addition to the treatment in the model group),and high concentration group(treated with 120 mg/L CH-I in addition to the treatment in the model group).SA-β-Gal staining was used to measure the senescence rate of PC12 cells in each group;flow cytometry was used to measure the apoptosis rate of cells in each group;Western blot was used to measure the relative expression levels of phosphorylated signal transducer and activator of transcription 3(p-STAT3)and telomerase reverse transcriptase(TERT)in PC12 cells,and PCR-ELISA was used to measure the activity of telomerase in each group.Results Compared with the control group,the model group had significant increases in the senescence rate and apoptosis rate of cells(t_(LSD)=27.03,17.77,P<0.05)and a significant reduction in telomerase activity(t_(LSD)=6.21,P<0.05).Compared with the model group,the low and high concentration groups had significant reductions in the senescence rate and apoptosis rate of SA-β-Gal cells(t_(LSD)=7.74-21.43,P<0.05)and significant increases in telomerase activity(t_(LSD)=2.01,2.49,P<0.05)and the relative expression levels of TERT and p-STAT3 in cells(t_(LSD)=2.21-6.03,P<0.05).Conclusion CH-Ⅰmay alleviate the senescence of PC12 cells induced by D-Gal by promoting STAT3 phosphorylation,upregula-ting the expression of TERT,and increasing telomerase activity.
