朝医方两仪煎颗粒制备工艺研究

摘  要:目的优化两仪煎颗粒最佳提取和制备工艺条件。方法采用正交试验设计,以毛蕊花糖苷、莫诺苷和马钱苷的含量和出膏率为考察指标,优化药材提取工艺;用两仪煎冻干粉为原料,以成型率、堆密度、休止角、溶化率和吸湿性为考察指标,对药粉与辅料的比例、润湿剂和颗粒干燥温度进行单因素考察,优选两仪煎颗粒的制备工艺。结果最佳提取工艺为加水8倍量,提取2次,每次1.5 h;最佳成型工艺为药粉∶糊精∶淀粉∶甘露醇=1∶1∶0.5∶0.5,以90%乙醇水溶液为润湿剂,70℃干燥,整粒。结论两仪煎颗粒制备及成型工艺简单可行,为该朝医方的开发利用提供实验依据。Objective To optimize the optimum extraction and preparation conditions of Liangyijian granules.Methods The orthogonal experiment design was used to optimize the extraction technology with the content and extract rate of verbascoside,morroniside and loganin and the extraction rate as indexes.Taking Liangyijian freeze-dried powder as raw material,taking molding rate,bulk density,angle of rest,solubility,and hygroscopicity as indexes,the proportion of auxiliary materials,wetting agent and drying temperature of granules were investigated by single factor.Results The optimal extraction conditions were as follows:adding 8 times of water,extracting twice for 1.5 h each time.The best molding process is that primary drug∶dextrin∶starch∶mannitol=1∶1∶0.5∶0.5,90%ethanol aqueous solution is used as wetting agent,drying at 70℃and granulated.Conclusion The preparation and molding process of Liangyijian granules is simple and feasible,which provides experimental basis for the development and utilization of Chinese Korean medical prescription.
