柴胡疏肝散化裁方加味江香薷对肥胖抑郁大鼠的药效实验研究

摘　　要：目的:在高脂饮食和慢性不可预见性温和应激法(CUMS)的基础上建立肥胖抑郁大鼠模型,并探讨柴胡疏肝散化裁方加味江香薷对肥胖抑郁大鼠的药效作用。方法:除正常组、抑郁组大鼠外先进行高脂饮食进行肥胖造模,再采用CUMS方法进行抑郁造模,进行行为学检测后,对肥胖抑郁模型组,低、中、高柴胡疏肝散加味江香薷组,肥胖抑郁与氟西汀组进行给药,其余组均给生理盐水,给药30 d后采集血液制备血清检测血脂、神经递质和肠道屏障及动力学指标,获取其粪便进行16S rDNA高通量测序技术获取实验数据进行分析。结果:肥胖抑郁大鼠在给予柴胡疏肝散化裁方加味江香薷后,行为学测试有所改善,血脂水平下降,胃及肠道屏障功能、HPA轴相关激素以及5-HT水平得到改善,肠道菌群的丰度增加,多样性变化增加。结论:通过持续的高脂饮食和CUMS建立肥胖抑郁模型,柴胡疏肝散化裁方加味江香薷可能通过调节肠道菌群的相对丰度达到治疗肥胖抑郁症的效果。Objective:To establish a model of obesity and depression in rats on the basis of high-fat diet and chronic unpredictable mild stimulation(CUMS),and to explore the pharmacodynamic effect of Chaihu Shugan Powder Modified Jiangxiangru on obesity and depression in rats.Methods:Obesity depression model was established by simulating high-fat diet and CUMS.Then,the obesity depression model group,low,medium and high Chaihu Shugan Powder Modified Jiangxiangru group and the obesity depression and fluoxetine group were administered.The other groups were given normal saline.After 30 days of administration,blood was collected and their feces were obtained for analysis by 16Sr DNA high-throughput sequencing technology.Results:After the obese and depressed rats were given Chaihu Shugan Powder Modified Jiangxiangru,the behavioral test was improved,the blood lipid level was decreased,the gastric and intestinal barrier function,HPA axis related hormones and 5-HT levels were improved,the abundance of gut flora was increased,and the diversity was increased.Conclusion:The obesity depression model was established by continuous high-fat diet and CUMS.Chaihu Shugan Powder Modified Jiangxiangru may achieve the effect of treating obesity depression by regulating the relative abundance of gut flora.
