摘 要:目的利用Box-Behnken响应面设计法结合熵权法优选经典名方“开心散”的提取工艺。方法采用高效液相色谱法同时测定开心散活性成分3,6′-二芥子酰基蔗糖、人参皂苷Rg1、β-细辛醚、茯苓酸的含量。基于多指标评价体系,结合提取液的出膏率和活性成分含量进行评分,通过单因素实验分别考察料液比、浸泡时间和回流提取时间对开心散提取工艺的影响。在此基础上,采用Box-Behnken响应面法设计实验,利用熵权法计算各指标权重系数并进行综合评分。以综合评分为评价指标,建立影响因素与综合评分之间的函数模型,预测并优选开心散的最佳提取工艺参数。最后,对优选后的提取工艺进行验证。结果熵权法计算结果表明,开心散中3,6′-二芥子酰基蔗糖、人参皂苷Rg1、β-细辛醚和茯苓酸的权重系数分别为0.1912,0.2025,0.1634和0.2954,出膏率的权重系数为0.1476。以料液比(A)、浸泡时间(B)、回流提取时间(C)为影响因素所建立的与综合评分之间的函数模型为:综合评分=98.69+2.86A-2.2B+1.33C-1.23AB-1.72AC-0.5575BC-9.67A2-9.28B2-11.86C2。根据模型预测,开心散的最佳提取工艺条件为料液比1∶10、浸泡40 min、回流提取1 h、提取2次。3批最佳提取工艺验证实验结果的综合评分平均为98.93,RSD为0.12%,综合评分实际值与预测值偏差在±1%以内。结论本研究所优选的开心散提取工艺稳定可靠,模型预测性好,为开心散的提取工艺及基准样品研究提供科学实验参考。Objective To optimize the extraction process of Kai-Xin-San(KXS)by Box-Behnken response surface design with entropy weight method.Methods The contents of KXS active ingredients 3,6′-disinapoyl sucrose,ginsenoside Rg1,β-asarone,and pachymic acid were simultaneously determined by high performance liquid chromatography.Based on a multi-criteria evaluation system,the paste rate and the active ingredient contents of the extract,scoring was performed.Single-factor experiments were conducted to investigate the effects of material-to-liquid ratio,soaking time and reflux extraction time on the extraction process of KXS.On this basis,a Box-Behnken response surface design was used to conduct experiments.The entropy weight method was applied to calculate the weight coefficients of each indicator and determine the comprehensive score.To predict and screen the best extraction process parameters for KXS,the comprehensive score was used as the evaluation criterion to establish a functional model between the influencing factors and the comprehensive score.Finally,the optimized extraction process was verified.Results The weight coefficients of 3,6′-disinapoyl Sucrose,ginsenoside Rg1,β-asarone,pachymic acid,and extraction rate determined by entropy weight method were 0.1912,0.2025,0.1634,0.2954,and 0.1476,respectively.The established function model was comprehensive score=98.69+2.86A-2.2B+1.33C-1.23AB-1.72AC-0.5575BC-9.67A2-9.28B2-11.86C2,with material-to-liquid ratio as factor A,soaking time as factor B,reflux extraction time as factor C.According to the model prediction,the optimal extraction conditions for KXS were as follows:a material-to-liquid ratio of 1∶10,a soaking duration of 40 min,reflux extraction for 1 h,and two extractions.The average comprehensive score of the three batches by optimal extraction process was 98.93,and RSD was 0.12%.The deviation between the actual value and the predicted value was within±1%.Conclusion The optimized extraction process is stable and reliable,and the model is predictive,which ca
