摘  要:目的探讨壮医药线点灸对树脂毒素诱导的带状疱疹后遗神经痛(postherpetic neuralgia,PHN)大鼠的治疗作用及机制。方法75只SD雄性大鼠,随机分为盐水组、模型组、点灸组、吡咯烷二硫代甲酸铵(pyrrolidinedithiocarbamate,PDTC)组、PDTC+点灸组,每组15只。除盐水组外,其余大鼠均腹腔注射树脂毒素建立PHN样大鼠模型。点灸组于造模成功后隔日予壮医药线点灸治疗,隔日点灸一次,10次为一疗程;PDTC组建模成功后予以腹腔注射核因子-κB(nuclear factor kappa-B,NF-κB)抑制剂PDTC 50 mg/kg,每日1次;PDTC+点灸组在建模成功后腹腔注射PDTC,并采用壮医药线点灸疗法治疗。定期测量各组大鼠行为学改变情况,同时对背根神经节组织进行实时荧光定量PCR、酶联免疫吸附法、RNA测序、蛋白印迹的检测及分析。结果壮医药线点灸降低了PHN大鼠背根神经节中肿瘤坏死因子-α、白细胞介素-1β和白细胞介素-6等炎症因子的mRNA和含量表达(均P<0.05)。RNA测序分析显示点灸组与模型组之间的差异基因在NF-κB信号通路显著富集,后续通过蛋白印迹的方法得到证实(均P<0.05)。与模型组相比,点灸组和PDTC组的p65、p-p65、NF-κB抑制蛋白(inhibitor of NF-κB,IκB)α和p-IκBα的蛋白表达水平显著降低(均P<0.05)。与点灸组相比,PDTC+点灸组的p65、p-p65、IκBα和p-IκBα的蛋白表达水平也显著降低(均P<0.05)。结论壮医药线点灸通过抑制NF-κB信号通路显著改善PHN大鼠的神经炎症。Objective To explore the therapeutic effect and mechanism of Zhuang medicine thread moxibustion on RTX induced postherpetic neuralgia(PHN)in rats.Methods 75 male SD rats were randomly divided into saline group,model group,moxibustion group,pyrrolidine dithiocarbamate(PDTC)group,and PDTC+moxibustion group.Except for the saline group,all other groups were intraperitoneally injected with resin toxin to establish the PHN like rat model.The moxibustion group was treated with Zhuang Medicine thread moxibustion every other day after successful modeling,once every other day,with 10 sessions as a course of treatment;After successful modeling of the PDTC group,intraperitoneal injection of NF-κB inhibitor PDTC 50 mg/kg once a day was administered;The PDTC+moxibustion group:After successful modeling,PDTC was injected into the abdominal cavity and treated with Zhuang medicine thread moxibustion therapy.Regularly measure the behavioral changes of rats in each group,and the method of RT-PCR,ELisa,RNA sequencing(RNA seq),Western blot(WB)were used to analysis the dorsal root ganglion tissue.Results Zhuang medicine thread moxibustion reduced the mRNA and content expression of inflammatory factors such as TNF-α,IL-1β,and IL-6 in the dorsal root ganglia of PHN rats(P<0.05).The result of RNA seq analysis showed that the difference between Zhuang medicine thread moxibustion and the model group were significantly enriched in the NF-κB signaling pathway,which was subsequently confirmed by Western blot(P<0.05).Compared with the model group,the protein expression levels of p65,p-p65,IκBα,and p-IκBαin the moxibustion group and the PDTC group were significantly decreased(P<0.05).Compared with the moxibustion group,the protein expression levels of p65,p-p65,IκBα,and p-IκBαin the PDTC+moxibustion group were significantly reduced(P<0.05).Conclusion Zhuang medicine thread moxibustion significantly improves neuroinflammation in PHN rats by inhibiting the NF-κB signaling pathway.
