云药七龙天对低压低氧大鼠肺血管重构的作用及机制研究

摘  要:目的探讨云药七龙天对低压低氧大鼠肺血管重构的保护作用及机制。方法将SD雄性大鼠随机分为3组:空白组、模型组、七龙天组,每组15只。除空白组外,造模前3天开始给药,七龙天组给予七龙天混悬液2 mL/只,剂量3.5 g/(kg·d);模型组予等体积蒸馏水灌胃,每日灌胃1次,连续给药3天。给药3天后采用低压氧仓构建低压低氧模型,连续72小时。造模完成后,肺组织苏木素—伊红染色观察形态学改变,测定各组大鼠血清白细胞介素(interleukin,IL)-1β水平;以实时荧光定量PCR、蛋白免疫印迹法检测肺组织中S100钙结合蛋白A9(S100 Calcium Binding Protein A9,S100A9)、IL-17b、趋化因子配体2[chemokine(C-C motif)ligand 2,Ccl2]、α-平滑肌激动蛋白(α-smooth muscle Actin,α-SMA)、基质金属蛋白酶-9(matrix metalloproteinase 9,MMP-9)、肿瘤坏死因子配体超家族成员9(tumor necrosis factor ligand superfamily member 9,Tnfsf9)、Fas凋亡抑制分子(Fas apoptotic inhibitory molecule,Faim)的mRNA与蛋白表达;免疫组化检测CD 68的蛋白表达。结果苏木素—伊红染色结果表明:与空白组对比,模型组大鼠肺血管出现明显的血管壁增厚;与模型组相比,七龙天组大鼠肺血管增厚程度有所改善。与空白组比较,模型组血清IL-1β显著升高(P<0.01)。与模型组相比,七龙天组IL-1β水平明显下降(P<0.01)。与空白组相比,模型组的S100A9、IL-17b、Ccl2、α-SMA、MMP-9、Tnfsf9、Faim的mRNA与蛋白表达均显著升高(P<0.01);与模型组相比,七龙天组S100A9、IL-17b、Ccl2、α-SMA、MMP-9、Tnfsf9、Faim的mRNA与蛋白表达均明显下降(P<0.01)。结论云药七龙天可降低低压低氧模型大鼠的炎症因子血清水平,降低肺组织炎症因子、趋化因子、血管重塑相关因子、细胞凋亡因子的表达水平,减少肺组织免疫细胞浸润,减轻肺组织炎症反应,从而改善低压低氧大鼠肺血管重构,其干预机制待进一步细胞水平验证。Objective To investigate the protective effect and mechanism of Qilongtian on pulmonary vascular remodeling in acute hypoxia-induced lung injury in rats.Methods SD male rats were randomly divided into 3 groups:blank group,model group,and Qilongtian group,with 15 rats in each group.Except for the blank group,in the Qilongtian group,2 mL of Qilongtian suspension at a dose of 3.5 g/(kg·d)was administered 3 days prior to modeling,the model group received an equal volume of distilled water,administered once daily for 3 consecutive days.Morphologic changes in lung tissue observed by HE staining,serum IL-1βlevel was measured,mRNA and protein expression of S100A9,IL-17b,α-SMA,MMP9,Ccl2,Tnfsf9,Faim in lung tissue were detected by RT-qPCR,Western blot,and protein expression of CD 68 was detected by immunohistochemistry.Results HE staining results showed that compared with the blank group,the pulmonary blood vessels of rats in the model group showed obvious thickening of the vessel wall;compared with the model group,the degree of thickening of the pulmonary blood vessels of rats in the Qilongtian group was improved;The model group has significantly higher serum IL-1βlevels than the blank group,and compared with the model group,the Qilongtian group could significantly reduce the level of IL-1β(P<0.01).The mRNA and protein expression of S100A9,IL-17b,Ccl2,α-SMA,MMP9,Tnfsf9,and Faim in the model group were all significantly elevated.Compared to the model group,the Qilongtian group could reduce the level of S100A9,IL-17b,Ccl2,α-SMA,MMP9,Tnfsf9 and Faim,IL-17b,Ccl2,Tnfsf9,and Faim mRNA and protein expression(P<0.01),while reducing macrophage and T cell recruitment.Conclusion Qilongtian may reduce serum levels of inflammatory factors,lower expression levels of inflammatory,chemotactic and apoptotic factors in lung tissue,reduce the immune cellinfiltrations,and alleviate the inflammatory response,thereby improving pulmonary vascular remodeling in rats under hypobaric hypoxia.Further cellular studies are needed to confirm th
