甘草干姜汤不同炮制品抗过敏性鼻炎的药效比较

摘　　要：目的通过分析比较甘草干姜汤不同炮制品对大鼠嗜碱性白血病细胞(rat basophil leukemia,RBL-2H3)脱颗粒及过敏性鼻炎模型小鼠的影响,考察甘草干姜汤不同炮制品抗过敏性鼻炎的药效。方法RBL-2H3细胞复苏传代后,分为生甘草干姜组、炙甘草干姜组、炙甘草炮姜组、炙甘草姜炭组,给药培养24小时,通过CCK-8法检测药物的毒性作用。取对数生长期细胞,设置给药组、模型组、空白组,模型组及空白组给予完全培养基,给药组加入相同剂量(0.4 mg/mL)的甘草干姜汤不同炮制品含药培养基,培养24小时。尔后加入浓度为80μg/mL的C48/80孵育,离心取上清,检测各组上清中β-氨基己糖胺酶含量以及白细胞介素4(interleukin-4,IL-4)含量,观察不同甘草干姜汤炮制品对C48/80诱导RBL-2H3细胞脱颗粒模型的影响。将48只SPF雌性BALB/C小鼠随机分为6组(空白组、模型组、阳性药组、生甘草干姜组、炙甘草干姜组、炙甘草姜炭组),每组8只。将卵清蛋白(ovalbumin,OVA)溶于生理盐水与同体积铝佐剂,在实验第1、7、14天以0.2 mL/只的剂量进行腹腔注射,空白组注射等量生理盐水。实验第22~32天,给药组以1%OVA滴鼻激发,20μL/(鼻孔·次),每日1次,连续10天。空白组小鼠以相同体积生理盐水滴鼻激发。滴鼻30分钟前灌胃给药,除空白组及模型组外,其余各给药组给予对应药物。末次滴鼻后观察10分钟内小鼠喷嚏、搔鼻次数。取小鼠鼻黏膜以及眼球血,进行鼻黏膜苏木素—伊红(hematoxylin-eosin,HE)染色及小鼠血清卵清蛋白特异性免疫球蛋白E(ovalbumin specific immunoglobulin E,OVA-sIgE)含量检测。结果细胞实验显示各炮制品组对RBL-2H3细胞均无活性抑制作用;各给药组可明显降低β-氨基己糖胺酶释放率(P<0.05)并且显著降低IL-4释放含量(P<0.01),其中炙甘草姜炭组的效果最好。小鼠行为学实验结果显示,与模型组相比,炙甘草姜炭组和炙甘草干�Objective To compare the effects of different processed products of Gancao Ganjiang Decoction on the degranulation of rat basophil leukemia(RBL-2H3)cells and allergic rhinitis model mice,and to investigate the anti-allergic rhinitis efficacy of different processed products of Gancao Ganjiang Decoction.Methods RBL-2H3 cells were divided into four groups:the raw Gancao+Ganjiang group,the burnt Gancao+Ganjiang group,the burnt Gancao+dried ginger group,the burnt Gancao+charcoal of ginger.After 24 hours of incubation,the toxicity of drugs was detected by CCK-8 method.The cells in logarithmic growth phase were divided into the administration group,the model group and the blank group.The model group and the blank group were given complete medium,and the administration group was added with the same dose(0.4 mg/mL)of different processed products of Gancao Ganjiang Decoction.After incubation with C48/80 at a concentration of 80μg/mL,the supernatant was centrifuged,and the content ofβ-hexosaminidase and interleukin-4(IL-4)in the supernatant of each group was detected to observe the effect of different processed products of Gancao Ganjiang Decoction on C48/80 induced RBL-2H3 cell degranulation model.Forty-eight specific pathogen-free female BALB/C mice were randomly divided into 6 groups(the blank group,the model group,the positive drug group,the raw Gancao+Ganjiang group,the burnt Gancao+Ganjiang group,the burnt Gancao+Charcoal of ginger),with 8 mice in each group.Ovalbumin(OVA)was dissolved in normal saline and the same volume of aluminum adjuvant,and the mice were intraperitoneally injected with 0.2mL OVA on the 1st,7th and 14th day of the experiment.The blank group was injected with the same volume of normal saline.From the 22th day to the 32th day,the mice in the control group were challenged with 1%OVA by nasal drops,20μL/(nostril·time),once a day for 10 consecutive days.Mice in the blank group were challenged by nasal drops of the same volume of normal saline.Except for the blank group and the model group,the othe
