辣木叶提取物通过缓解氧化应激对急性高原低氧暴露小鼠心肺损伤的减轻作用研究

摘  要:目的研究辣木叶提取物是否通过缓解氧化应激来减轻急性高原低氧暴露小鼠的心肺损伤。方法40只雄性昆明小鼠,随机分为常氧对照组(NC)、低氧对照组(HC)、辣木叶提取物低剂量组(LMLE)、辣木叶提取物高剂量组(HMLE),每组10只。LMLE和HMLE组分别给予1000、2000 mg/kg的辣木叶提取物混悬液灌胃,NC和HC组灌胃蒸馏水。以低压氧舱模拟6000 m海拔,HC组、LMLE、HMLE组每日低氧暴露23 h,NC组于常氧环境中饲养,持续3 d。处理结束后,麻醉取血,随后处死动物,取心、肺组织,测定血清及组织氧化应激相关指标、血清LDH和CK-MB水平,观察肺组织病理学变化。结果与NC组比较,暴露结束后HC组小鼠体重明显下降,肺泡壁增厚,肺间质充血;小鼠血清、肺组织及心肌组织SOD和GSH-Px活性、T-AOC明显降低,MDA水平明显升高,差异有统计学意义(P<0.05);血清LDH及CK-MB水平明显升高,差异有统计学意义(P<0.05)。与HC组比较,辣木叶提取物组小鼠体重下降程度较少,肺组织损伤减轻;小鼠血清、肺组织及心肌组织SOD和GSH-Px活性、T-AOC明显升高,MDA水平明显减少,差异有统计学意义(P<0.05);血清LDH和CK-MB水平明显降低,差异有统计学意义(P<0.05)。结论辣木叶提取物能够减轻急性低氧暴露小鼠肺组织和心肌组织损伤,这可能与辣木叶提取物降低机体的氧化应激损伤有关。Objective To investigate whether Moringa oleifera leaf extract(MLE)can alleviate the cardiopulmonary injury in mice exposed to acute altitude hypoxia by reducing oxidative stress.Methods Forty male Kunming mice were randomly divided into normoxia control(NC)group,hypoxia control(HC)group,Moringa oleifera leaf extract low-dose group(LMLE)and high-dose Moringa oleifera leaf extract group(HMLE),10 mice in each group.The LMLE and HMLE groups were given 1000 mg/kg and 2000 mg/kg MLE suspension respectively,through gavage,and the NC and HC groups were given distilled water.A low-pressure oxygen chamber was used to simulate the altitude of 6000 m.The HC group,LMLE group and HMLE group were exposed to hypoxia for 23 h every day,and the NC group was fed in normoxia environment for 3 days.After the treatment,the blood was taken under anesthesia.Then the animals were sacrificed and the heart and lung tissues were collected.The oxidative stress related indexes in serum and tissues,and LDH and CK-MB levels in serum were measured and histopathological examination of lung tissue were done.Results Compared with the NC group,the weight of mice in HC group was decreased significantly,the alveolar wall was thickened,and pulmonary congestion was observed.The activities of SOD and GSH-Px,and T-AOC in serum,lung tissue and myocardial tissue significantly decreased(P<0.05),while the content of MDA significantly increased(P<0.05).Serum LDH and CK-MB levels significantly increased(P<0.05).Compared with the HC group,the weight loss of mice in MLE group was less,and lung tissue injury was alleviated.The activities of SOD and GSH-Px,and T-AOC in serum,lung tissue and myocardial tissue of mice significantly increased(P<0.05),while the content of MDA significantly decreased(P<0.05).Serum LDH and CKMB levels significantly decreased(P<0.05).Conclusion MLE can reduce the damage of lung and myocardia in mice exposed to acute hypoxia,which may be related to the reduction of oxidative stress damage by MLE.
