摘  要:磷脂酰肌醇3-激酶/蛋白激酶B(phosphatidylinositol 3-kinase/protein kinase B,PI3K/AKT)信号通路参与调节细胞的生存、生长、增殖及凋亡等过程,与哮喘的发生与发展关系密切。研究表明,中药单体通过调控PI3K/AKT信号通路,可从抑制炎症反应、氧化应激、自噬等多方面,干预哮喘气道炎症、气道黏液分泌、气道重塑等过程。虽然目前中药单体基于PI3K/AKT信号通路抑制哮喘发展的相关研究已深入到分子与基因水平,但仍然存在许多不足之处:(1)关于中药单体靶向调控PI3K/AKT通路的具体作用机制研究尚未成熟,单项实验通常以PI3K/AKT信号通路上游或下游较为单一的靶点作为观察指标,未能明确不同靶点交互的作用机制。(2)大多数研究集中在体外细胞实验与体内动物实验阶段,缺乏相关临床试验研究的验证与支持,此类单体成分应用于人体的有效性与安全性有待进一步验证。因此,在未来的研究中,可以在现阶段研究基础上开展体内外实验研究,筛选出机制明确并精准靶向PI3K/AKT信号通路的中药单体,同时推进临床研究的开展,增强基础研究的临床转化,为新药开发奠定基础。The phosphatidylinositol 3-kinase/protein kinase B(PI3K/AKT)signaling pathway is involved in the regulation of cell survival,growth,proliferation,and apoptosis,which is closely related to the occurrence and development of asthma.Studies have shown that Chinese medicine monomers can intervene in asthma airway inflammation,airway mucus secretion,airway remodeling and other processes by regulating the PI3K/AKT pathway and inhibiting inflammatory response,oxidative stress,autophagy,and other aspects.Although the research on the inhibition of asthma based on the PI3K/AKT signaling pathway by traditional Chinese medicine monomers has been deepened to the molecular and genetic levels,there are still many shortcomings:(1)The research on the specific mechanism of traditional Chinese medicine monomers in targeting and regulating PI3K/AKT has not yet matured,and a single experiment is usually based on the observation of a single target upstream or downstream of PI3K/AKT signaling pathway as an indicator.Failed to clarify the mechanism of interaction between different targets.Most of the studies have focused on in vitro cellular experiments and in vivo animal experiments and lack the verification and support of relevant clinical trials,so the effectiveness and safety of these monomer components applied to humans need to be further verified.Therefore,in the future,we can carry out in vitro and in vivo experimental studies based on the current research,screen out the Chinese medicine monomers with clear mechanism and precise targeting of PI3K/AKT signaling pathway,and promote the clinical research to enhance the clinical translation of the basic research,to lay the foundation for the development of new drugs.
