摘 要:目的建立高效液相色谱(HPLC)多波长法同时测定安体威颗粒中8种成分的含量。方法采用Venusil XBP Polar-Phenyl色谱柱(4.6 mm×250 mm,5μm),以0.1%磷酸水(A)-甲醇(B)-乙腈(C)为流动相梯度洗脱,检测波长0~26 min:210 nm,26~34 min:320 nm,34~60 min:240 nm;流速1.0 mL/min;柱温30℃;进样量10μL。结果盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷、葛根素、大豆苷、大豆苷元、甘草酸铵、6-姜辣素分别在12.81~205.00、10.57~169.16、30.58~489.26、100.74~2014.85、20.09~321.44、2.09~41.83、10.13~162.07、4.20~63.42μg/mL与峰面积线性关系良好(r=0.9999、0.9999、0.9999、1.0000、0.9999、1.0000、1.0000、0.9998);平均回收率[相对标准偏差(RSD)]分别为98.32%(1.33%)、103.45%(0.88%)、105.10%(1.09%)、102.04%(1.35%)、99.49%(2.14%)、99.54%(2.30%)、97.90%(1.53%)、106.27%(2.98%)。结论该方法可以同时测定安体威颗粒中8种成分的含量,为其质量标准的提升奠定了基础。Objective To establish a multi-wavelength high performance liquid chromatography(HPLC)method for the simultaneous determination of the contents of eight components in Antiwei granules.Methods The chromatographic column was a Venusil XBP Polar-Phenyl column(4.6 mm×250 mm,5μm),and gradient elution was conducted with 0.1%phosphoric acid(A)-methanol(B)-acetonitrile(C)as the mobile phase.The detection wavelength was set at 210 nm(0~26 min),at 320 nm(26~34 min),at 240 nm(34~60 min).The flow rate was 1.0 mL/min,the column temperature was 30℃and the injection volume was 10μL.Results The linear relationships between the peak areas and concentrations of ephedrine hydrochloride,pseudoephedrine hydrochloride,amygdalin,puerarin,daidzin,daidzein,glycyrrhizic acid ammonium,and 6-gingerol were good in the ranges of 12.81~205.00,10.57~169.16,30.58~489.26,100.74~2014.85,20.09~321.44,2.09~41.83,10.13~162.07,and 4.20~63.42μg/mL,respectively(r=0.9999,0.9999,0.9999,1.0000,0.9999,1.0000,1.0000,0.9998).The average recovery rates[relative standard deviation(RSD)]were 98.32%(1.33%),103.45%(0.88%),105.10%(1.09%),102.04%(1.35%),99.49%(2.14%),99.54%(2.30%),97.90%(1.53%),and 106.27%(2.98%).Conclusion This method could simultaneously determine the content of eight components in Antiwei granules,and provided a foundation for the improvement of the quality standard.
