摘  要:目的研究苁蓉总苷对功能性便秘大鼠肠黏膜损伤、Cajal间质细胞(ICC)、水通道蛋白(AQP)及胃肠动力学的影响。方法选取52只健康大鼠为实验动物,随机抽取10只大鼠作为健康组,剩余42只大鼠建立功能性便秘模型。模型建立成功后随机分为模型组、苁蓉总苷组(模型+苁蓉总苷10 mg/kg)与药物对照组(模型+西沙必利0.1 mL/10 g),每组14只。采用酶联免疫吸附试验(ELISA)检测各组大鼠胃肠动力学相关指标[5-羟色胺(5-HT)、一氧化氮(NO)、肠内P物质(SP)],苏木素-伊红染色检测大鼠结肠组织病理形态变化,定量聚合酶链反应检测各组大鼠结肠组织toll样受体(TLR)2mRNA、TLR4mRNA、闭锁小带蛋白1-(Claudin-1)mRNA水平,免疫组化法检测结肠组织中ICC标志物C-kit,蛋白质印迹法检测各组大鼠结肠组织中AQP相关指标。结果与健康组比较,模型组、苁蓉总苷组、药物对照组SP、VIP、C-kit平均光密度值(AO值)、AQP4、AQP9、Claudin-1mRNA水平降低,NO、5-HT、TLR2mRNA、TLR4mRNA、AQP2、AQP3水平升高,差异均有统计学意义(P<0.05);与模型组比较,苁蓉总苷组与药物对照组SP、VIP、C-kit AO值、AQP4、AQP9、Claudin-1mRNA水平升高,NO、5-HT、TLR2mRNA、TLR4mRNA、AQP2、AQP3水平降低,差异均有统计学意义(P<0.05);苁蓉总苷组与药物对照组各项指标水平比较,差异均无统计学意义(P>0.05)。健康组大鼠结肠黏膜完整、腺体丰富且排列有序;模型组大鼠结肠黏膜的完整性被破坏,腺体萎缩、腺泡减少且杂乱无章;苁蓉总苷组与药物对照组大鼠病理变化相对模型组有所改善。结论苁蓉总苷可能是通过调控肠神经递质、AQP、ICC改善肠黏膜损伤,从而发挥了治疗功能性便秘的作用。Objective To study the effects of total Cistanche glycosides on intestinal mucosal injury,Cajal interstitial cells(ICC),aquaporin(AQP)and gastrointestinal dynamics in rats with functional constipation.Methods Among 52 healthy rats,10 rats were randomly selected as the healthy group,and the remaining 42 rats were established as the functional constipation model.After successful model establishment,the rats were randomly divided into model group,total Cistanche glycosides group(model+total Cistanche glycosides 10 mg/kg)and drug control group(model+cisapride 0.1 mL/10 g),with 14 rats in each group.Enzyme linked immunosorbent assay(ELISA)was used to detect gastrointestinal motility related indicators 5-hydroxytryptamine(5-HT),nitric oxide(NO),intestinal substance P(SP)],asoactive intestinal Peptide(VIP).Hematoxylin-Eosin staining was used to detect the pathological changes of colon tissue.qPCR was used to detect the expressions of TLR2mRNA,TLR4mRNA and Claudin-1mRNA in colon tissue.Immunohistochemistry was used to detect the ICC marker C-kit in colon tissue.Western blotting was used to detect AQP-related indexes in colonic tissue of rats in each group.Results Compared with the healthy group,the levels of SP,VIP,C-kit AO,AQP4,AQP9 and claudin-1mRNA decreased in the model group,total Cistanche glycosides group and drug control group,and the levels of NO,5-HT,TLR2mRNA,TLR4mRNA,AQP2 and AQP3 increased,the differences were significant(P<0.05).Compared with the model group,the levels of SP,VIP,C-kit AO,AQP4,AQP9 and claudin-1mRNA increased significantly in the total Cistanche glycosides group and the drug control group(P<0.05),and the levels of NO,5-HT,TLR2 mRNA,TLR4 mRNA,AQP2 and AQP3 decreased significantly in the total Cistanche glycosides group and the drug control group(P<0.05).There was no statistical significance in the levels of each index between the total Cistanche glycosides group and the drug control group(P>0.05).The colonic mucosa of healthy rats was intact,and the glands were abundant and arranged in an orde
