摘 要:目的 探讨Polo样激酶1(PLK1)基因在肺腺癌(LUAD)中的表达、临床价值及与LUAD发生、发展的关系。方法 GEO数据库中筛选LUAD的差异表达基因并根据相关性强度确定核心基因。利用Oncomine数据库分析核心基因在LUAD中的表达变化。Kaplan-Meier生存分析用于评估PLK1表达水平与总生存期(OS)的相关性;logsitic回归模型用于评价PLK1表达水平与不同临床病理学参数的相关性;Cox回归模型用于探讨影响LUAD患者生存的预后因素。结果 PLK1在LUAD中显著上调,并与临床分期、T期、N期、M期及性别显著相关;Kaplan-Meier分析及单变量Cox回归均显示,PLK1的高表达与OS相关,多变量分析确认其为独立预后因子。结论 PLK1可能是LUAD的一个潜在的独立预后因素。Objective To investigate the expression,clinical value,and relationship between Polo-like kinase 1(PLK1)gene and the occurrence and development of lung adenocarcinoma(LUAD).Methods Differentially expressed genes of LUAD selected from GEO database and core genes based on correlation strength were determined.The expression changes of core genes in LUAD were analyzed using the Oncomine database.Kaplan Meier survival analysis was used to evaluate the correlation between PLK1 expression levels and overall survival(OS).The logistic regression model was used to evaluate the correlation between PLK1 expression levels and different clinical pathological parameters.Cox regression model was used to investigate prognostic factors impacting the survival of patients with LUAD.Results PLK1 was significantly upregulated in LUAD and significantly correlated with clinical stage,T stage,N stage,M stage,and gender;Kaplan-Meier analysis and univariate Cox regression both showed that high expression of PLK1 was associated with OS,and multivariate analysis confirmed it as an independent prognostic factor.Conclusion PLK1 may be a potential independent prognostic factor for LUAD.
