摘  要:目的对微小核糖核酸(microRNA,miRNA)-27a的单核苷酸多态性rs895819与2型糖尿病相关性进行Meta分析。方法通过PubMed、Embase、万方、CNKI、维普网检索有关miRNA-27a rs895919与糖尿病相关性的病例对照研究,检索日期为建库至2024年1月7日,由两名研究者根据纽卡斯尔-渥太华量表对纳入的文献进行质量评价。最终共纳入10篇文献,病例组4139例,对照组4171例。采用Stata 12进行数据分析,用优势比、95%可信区间计算rs895819各遗传模型与糖尿病的相关性,并对种族、糖尿病分型、样本量进行亚组分析。结果miRNA-27a rs895819的基因位点和基因型在糖尿病和健康对照两组人群中的分布频率比较,差异均无统计学意义(P均>0.05)。以种族进行分层分析发现,rs895819的突变位点C vs T、纯合子基因型CC vs TT、显性模型CT+CC vs TT和隐性模型CC vs CT+TT在高加索糖尿病和健康者人群的比较中,差异有统计学意义(P均<0.05);而rs895819的各遗传模型在亚洲和罗马尼亚两组人群中分布频率比较,差异均无统计学意义(P均>0.05)。以2型糖尿病和妊娠期糖尿病进行分层分析以及样本量≥1000和<1000进行分层分析时发现,该rs895819的各遗传模型在两组人群中的分布频率比较,差异无统计学意义(P>0.05)。结论研究表明miRNA-27a的rs895819突变可能降低高加索人群2型糖尿病的患病风险,但由于基因多态性、种族和地域的差异,未来需要在更大的样本人群中分析此突变位点与糖尿病的关联。Objective To conduct a Meta analysis of the relationship between single nucleotide polymorphism rs895819 of microRNA(miRNA)-27a and type 2 diabetes mellitus.Methods A comprehensive search was conducted inPubMed,Embase,Wanfang,and CNKI databases.The search date is from the establishment of the database to January 7,2024.The quality of the included literature was evaluated independently by two researchers according to the Newcastle-Ottawa Scale.Eventually we included 10 documents with a total of 4139 cases in the case group and 4171 cases in the control group.Stata 12 was used for data analysis and odds ratio,95%confidence interval were calculated to determine the correlation between each genetic model of rs895819 and diabetes mellitus.Subgroup analysis was performed based on race,diabetes type,and sample size.Results The results showed that the frequency distribution of the gene locus and genotype of miRNA-27a rs895819 in the diabetes mellitus and healthy control groups were compared,and the difference were not statistically significant(both P>0.05).Upon stratified analysis by race,it was found that the differences between mutation site C vs T,the homozygous genotype CC vs TT,the dominant model CT+CC vs TT,and the recessive model CC vs CT+TT in diabetes mellitus and healthy individuals in the Caucasian population were statistically significant(all P<0.05).However,the distribution frequencies of the various genetic models of rs895819 in the Asian and Romanian populations were compared,and the differences were not statistically significant(all P>0.05).Upon stratified analysis by type 2 diabetes mellitus and gestational diabetes mellitus,and upon stratified analysis by sample size≥1000 and<1000,it was found that the distribution frequencies of the various genetic models of rs895819 in the two populations was compared,and the difference was not statistically significant(P>0.05).Conclusion Our study suggests that the rs895819 mutation in miRNA-27a may lower the risk of type 2 diabetes mellitus in Caucasian population
