青蒿外植体灭菌处理方法的研究

摘　　要：[目的]建立青蒿外植体消毒、组织培养快繁体系。[方法]研究了75%乙醇、0.1%汞-吐温在不同消毒时间对青蒿嫩茎的灭菌效果,不同外植体的消毒效果对比以及通过在基础培养基添加PPM抗菌剂对外植体的灭菌影响。[结果]75%乙醇漂洗45 s、0.1%氯化汞-吐温浸泡消毒5.5 min,灭菌效果最佳;培养基添加0.1%PPM抗菌剂能进一步减低染菌率,提升灭菌效果,此时外植体嫩茎死亡率为40.0%,染菌率为32.0%,发芽率为32.0%。[结论]外植体中,嫩茎比嫩叶易培育,染菌率低,存活率较高。[Objective]The aim was to establish a rapid propagation system for Artemisia annua through tissue culture.[Method]The sterilization effects of 75%ethanol and 0.1%mercuric chloride-tween for different durations on the disinfection of tender stems of A.annua were studied.The comparative sterilization effects of different explants and the impact of adding PPM antimicrobial agents to the basic culture medium on explant sterilization were also investigated.[Result]The results indicated that rinsing with 75%ethanol for 45 seconds followed by a 5-minute and 30-second soak in 0.1%mercuric chloride-tween solution provided the best sterilization effect.The addition of 0.1%PPM antimicrobial agent to the culture medium further reduced the contamination rate and enhanced the sterilization effect,resulting in a 40.0%mortality rate of the tender stem explants,a 32.0%contamination rate,and a 32.0%germination rate.[Conclusion]Among the explants,tender stems were found to be easier to culture than tender leaves,with a lower contamination rate,higher survival rate,and reduced complications during the cultivation process.
