摘 要:目的探讨黄连素对幽门螺杆菌(Hp)感染模型小鼠胃内环境的影响。方法BALB/c雄性SPF级小鼠40只,随机分为正常组、正常+黄连素组、模型组、模型+黄连素组,各10只。以HpSS1菌液灌胃建立Hp感染小鼠模型。正常+黄连素组和模型+黄连素组小鼠予黄连素混悬液[60 mg/(kg·d)]灌胃治疗,连续给药4周。pH试纸法测定各组小鼠胃液pH值,酶联免疫吸附试验检测小鼠胃黏膜组织胃蛋白酶原、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、白细胞介素-10(IL-10)水平,苏木精-伊红(HE)染色观察小鼠胃黏膜病理变化。结果与正常组比较,模型组小鼠胃液pH值显著下降,胃黏膜组织胃蛋白酶原、IL-6、IL-8、IL-10水平显著升高(P<0.05或P<0.01)。与模型组比较,模型+黄连素组小鼠胃液pH值显著上升[(4.83±1.03)比(2.67±0.78),P<0.01],胃黏膜组织胃蛋白酶原[(3.74±0.91)pg/mL比(5.18±1.19)pg/mL,P<0.01]、IL-6[(7.27±1.53)pg/mL比(15.12±4.37)pg/mL,P<0.01]、IL-8[(27.56±4.31)pg/mL比(42.36±5.12)pg/mL,P<0.01]、IL-10[(8.32±1.78)pg/mL比(16.32±5.34)pg/mL,P<0.01]水平显著降低。病理结果显示,正常组和正常+黄连素组小鼠胃黏膜及黏膜下层结构清晰,腺体完整,未见病理改变;模型组小鼠胃黏膜结构破坏,可见大量炎症细胞浸润;而模型+黄连素组小鼠胃黏膜结构尚完整,只见少量炎症细胞浸润。结论黄连素可缓解Hp感染小鼠胃黏膜炎症反应,改善胃内环境。Objective To investigate the effects of berberine on the gastric environment in Helicobacter pylori(Hp)-infected mouse models.Methods Forty male BALB/c SPF-grade mice were randomly divided into four groups:the normal group,normal+berberine group,Hp-infected model group,and Hp-infected+berberine group,with 10 mice in each group.The Hp-infected mouse model was established by gavage with HpSS1 bacterial suspension.The mice in the normal+berberine group and Hp-infected+berberine group were treated with berberine suspension at a dose of 60 mg/(kg·d)by gavage for a continuous period of 4 weeks.The pH value of gastric juice in each group was measured using pH test paper.The levels of pepsinogen,interleukin-6(IL-6),interleukin-8(IL-8),and interleukin-10(IL-10)in the gastric mucosa tissues were detected using enzyme-linked immunosorbent assay.The pathological changes in the gastric mucosa were observed using hematoxylin-eosin staining.Results Compared with the normal group,the Hp-infected model group showed significantly decreased pH value of gastric juice and increased levels of pepsinogen,IL-6,IL-8,and IL-10 in the gastric mucosa tissues(P<0.05 or P<0.01).Compared with the Hp-infected model group,the Hp-infected+berberine group exhibited significantly increased pH value of gastric juice[(4.83±1.03)vs.(2.67±0.78),P<0.01],and significantly decreased levels of pepsinogen[(3.74±0.91)pg/mL vs.(5.18±1.19)pg/mL,P<0.01],IL-6[(7.27±1.53)pg/mL vs.(15.12±4.37)pg/mL,P<0.01],IL-8[(27.56±4.31)pg/mL vs.(42.36±5.12)pg/mL,P<0.01],and IL-10[(8.32±1.78)pg/mL vs.(16.32±5.34)pg/mL,P<0.01].Pathological results showed that the gastric mucosal and submucosal structures were clear in the normal group and the normal berberine group,with intact glands and no pathological changes.In contrast,the Hp-infected model group exhibited disrupted gastric mucosal structures with extensive inflammatory cell infiltration.The Hp-infected+berberine group showed relatively intact gastric mucosal structure with only mild inflammatory cell infiltration
