摘  要:用40%乙醇灌胃大鼠(剂量为15 mL·kg^(-1)),诱导肝损伤模型,用蜜环菌(Armillaria mellea)多糖(低、高剂量组分别命名为AMP-L、AMP-H,剂量为100、400 mg·kg^(-1))灌胃大鼠,4周后测定大鼠肝脏指数,血清天门冬氨酸氨基转移酶(AST)、γ-谷氨酰转移酶(GGT)、丙氨酸氨基转移酶(ALT)、超氧化物歧化酶(SOD)、超氧化物歧化酶2(SOD2)活性,清蛋白(ALB)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、丙二醛(MDA)含量;通过苏木素-伊红(HE)染色观察肝脏组织病理变化;采用Western blot方法测定核因子相关因子2(Nrf2)、血红素加氧酶-1(HO-1)、沉默信息调节因子3(Sirt3)和乙酰化超氧化物歧化酶2(Ac-SOD2)表达量。结果表明:与模型组相比,AMP-H的肝脏指数显著降低;AMP-L和AMP-H的ALT、AST、GGT活性显著降低,ALB含量显著增加;AMP-H的血清TG、TC含量极显著降低;AMP-L和AMP-H的血清LDL-C含量极显著降低,HDL-C含量极显著增加;AMP-H的SOD、SOD2活性显著增加;AMP-L和AMP-H的MDA含量显著降低,Nrf2、HO-1和Sirt3表达量极显著增加,Ac-SOD2表达量极显著降低,表明蜜环菌多糖对酒精性肝损伤具有保护作用。研究结果可为利用蜜环菌多糖缓解肝损伤提供参考。To study the protective effects of Armillaria mellea polysaccharide(AMP) on alcohol liver injury in rats, a liver injury model(MOD) was established by intragastrically administering 40%(V:V) ethanol to rats at a dose of 15 mL·kg^(-1) for four weeks. The model rats were then randomly divided into two groups: low-dose AMP group(AMP-L) at 100 mg·kg^(-1) and high-dose AMP group(AMP-H) at 400 mg·kg^(-1). After four weeks of treatment, rats in different groups were measured for liver index, activities of aspartate aminotransferase(AST), γ-glutamyltranspeptidase(GGT), alanine transaminase(ALT), superoxide dismutase(SOD), superoxide dismutase 2(SOD2), and levels of albumin(ALB), total cholesterol(TC), triglycerides(TG), low density lipoprotein cholesterol(LDL-C), high density lipoprotein cholesterol(HDL-C), and malondialdehyde(MDA).Morphological changes of liver tissue were observed through hematoxylin-eosin(HE) staining. Western blot analysis was used to determine the expression levels of nuclear factor-erythroid 2-related factor(Nrf2), heme oxygenase 1(HO-1), silent information regulator 3(Sirt3), and acetylated superoxide dismutase 2(Ac-SOD2)in liver tissue. The results showed that AMP ameliorated pathological damage in liver tissue with significantly decreased liver index in AMP-H, significantly decreased activities of ALT, AST and GGT in both AMP groups,significantly increased activities of SOD and SOD2 in AMP-H group, significantly increased ALB level in both AMP groups, extremely significant decreases of serum TG and TC levels in AMP-H group, extremely significant decrease of serum LDL-C in both AMP groups, extremely significant increase of serum HDL-C in both AMP groups, significantly decreased MDA level in both AMP groups, extremely significantly increased expression levels of Nrf2, HO-1, Sirt3, and extremely significantly decreased expression level of Ac-SOD2 in both AMP groups compared with MOD.
