摘 要:目的探讨艾片联合甘草查尔酮A(licorice chalcone A,LCA)对视网膜母细胞瘤Y79细胞增殖和侵袭的影响及其机制。方法2023年4—10月在牡丹江医科大学将体外培养的Y79细胞分为4组,包括空白对照组、艾片组、LCA组和艾片+LCA组。细胞计数试剂盒-8(cell counting kit-8,CCK-8)法测细胞增殖能力,Transwell小室法检测细胞侵袭能力,2,7-二氯荧光素二乙酸酯(2′,7′-dichlorodihydrofluorescein diacetate,DCFH-DA)荧光探针法检测胞内活性氧(reactive oxygen species,ROS)水平,实时荧光定量聚合酶链反应检测细胞周期蛋白D1(cyclin D1)和基质金属蛋白酶9(matrix metalloprotein 9,MMP-9)mRNA表达水平,蛋白印迹法检测缺氧诱导因子-1α(hypoxia-inducible factor 1,HIF-1α)和血管内皮生长因子(vascular endothlial growth factor,VEGF)蛋白的表达水平。结果与空白对照组比较,艾片组、LCA组、艾片+LCA组增殖活力、穿膜细胞数、Cyclin D1 mRNA、MMP-9 mRNA、HIF-1α蛋白和VEGF蛋白表达水平降低,差异有统计学意义(P均<0.05)。与艾片组、LCA组比较,艾片+LCA组增殖活力、穿膜细胞数、Cyclin D1 mRNA、MMP-9 mRNA、HIF-1α蛋白和VEGF蛋白表达水平降低,差异有统计学意义(P均<0.05)。与空白对照组比较,艾片组、LCA组、艾片+LCA组ROS水平升高,差异有统计学意义(P均<0.05);与艾片组、LCA组比较,艾片+LCA组ROS水平升高,差异有统计学意义(P均<0.05)。结论艾片联合LCA可抑制视网膜母细胞瘤Y79细胞的增殖和侵袭,其作用机制可能与上调ROS水平,下调Cyclin D1 mRNA、MMP-9 mRNA、HIF-1α蛋白和VEGF蛋白表达水平有关。Objective To investigate the effect of Aipian combined with licorice chalcone A(LCA)on the proliferation and invasion of retinoblastoma Y79 cells and its mechanism.Methods From April to October 2023,in Mudanjiang Medical University,Y79 cells cultured in vitro were divided into four groups,including blank control group,Aipian group,LCA group and Aipian+LCA group.Cell proliferation was measured by cell counting kit-8(CCK-8)method.Cell invasion was detected by Transwell chamber method.The level of intracellular reactive oxygen species(ROS)was detected by 2',7'-dichlorodihydrofluorescein diacetate(DCFH-DA)fluorescent probe method.The mRNA expression levels of cyclin D1 and matrix metalloprotein 9(MMP-9)were detected by real-time fluorescence quantitative poly-merase chain reaction.The protein expression levels of hypoxia-inducible factor 1(HIF-1α)and vascular endothelial growth factor(VEGF)were detected by Western blotting.Results Compared with the blank control group,the prolifera-tion activity,the number of transmembrane cells,the expression levels of Cyclin D1 mRNA,MMP-9 mRNA,HIF-1αprotein and VEGF protein in the Aipian group,LCA group and Aipian+LCA group were decreased,and the differ-ences were statistically significant(all P<0.05).Compared with Aipian group and LCA group,the proliferation activity,the number of transmembrane cells,the expression levels of Cyclin D1 mRNA,MMP-9 mRNA,HIF-1αprotein and VEGF protein in Aipian+LCA group were decreased,and the differences were statistically significant(all P<0.05).Compared with the blank control group,the ROS level of Aipian group,LCA group and Aipian+LCA group increased,and the difference was statistically significant(all P<0.05).Compared with the Aipian group and the LCA group,the ROS level in the Aipian+LCA group was increased,and the difference was statistically significant(both P<0.05).Conclusion Aipian combined with LCA can inhibit the proliferation and invasion of retinoblastoma Y79 cells,and its mechanism may be related to up-regulation of ROS level and dow
