摘  要:目的探究磷脂酰肌醇3-激酶(phosphoinositide 3-kinase,PI3K)/蛋白激酶B(protein kinase B,AKT)/哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)信号通路及其相关的细胞凋亡因子在缺血性脑卒中相关的神经元凋亡中的作用,为缺血性脑卒中的防治提供新的靶点。方法2023年1月—2024年5月,齐齐哈尔中心实验室购买HFK生物技术有限公司(北京,中国)的C57/B6L雄性小鼠20只,按随机数字表法分研究组和对照组,每组10只。构建体内大脑中动脉阻塞再灌注模型,提取脑组织,HE染色明确组织损伤严重程度。通过蛋白免疫印迹实验等检测细胞凋亡通路的相关因子。结果蛋白印迹实验的结果显示,研究组的PI3K、Akt、mTOR的表达分别为(0.75±0.03)mol/L、(0.65±0.35)mol/L、(0.65±0.39)mol/L,高于对照组的(0.27±0.06)mol/L、(0.38±0.36)mol/L、(0.47±0.76)mol/L;研究组的线粒体核糖体蛋白S9(mitochondrial ribosomal protein S9,MRPS9)表达为(0.14±0.08)mol/L,低于对照组的(0.50±0.10)mol/L,差异有统计学意义(t=41.355、48.614、43.587、32.561,P均<0.05);研究组Bax、caspase3的基因表达高于对照组,Bcl-2基因表达低于对照组,差异有统计学意义(P均<0.05)。实时荧光定量聚合酶链式反应的结果显示,研究组PI3K、Akt、mTOR的表达高于对照组,MRPS9的表达低于对照组,研究组Bax、caspase3的基因表达高于对照组,研究组Bcl-2基因表达低于对照组,差异有统计学意义(P均<0.05)。结论PI3K/Akt/mTOR参与神经细胞凋亡过程,在缺血性脑卒中的发展中发挥重要作用。Objective To explore the role of phosphoinositide 3-kinase(PI3K)/protein kinase B(AKT)/mammalian target of rapamycin(mTOR)signaling pathway and its related apoptotic factors in neuronal apoptosis associated with ischemic stroke and to provide new targets for the prevention and treatment of ischemic stroke.Methods From January 2023 to May 2024,Qiqihar Central Laboratory purchased twenty C57/B6L male mice from HFK Biotechnology Co Ltd(Beijing,China)and divided them into research group and control group according to randomized numerical table method,ten mice in each group.An in vivo middle cerebral artery occlusion-reperfusion model was constructed,brain tissues were extracted,and the severity of tissue damage was clarified by HE staining.Relevant factors of apoptosis pathway were detected by protein immunoblotting experiments.Results The results of protein blotting experiments showed that the expression of PI3K,Akt,and mTOR in the study group were(0.75±0.03)mol/L,(0.65±0.35)mol/L,(0.65±0.39)mol/L,respectively,which were higher than(0.27±0.06)mol/L,(0.38±0.36)mol/L and(0.47±0.76)mol/L in the control group;the expression of MRPS9 in the study group was(0.14±0.08)mol/L,which was lower than(0.50±0.08)mol/L in the control group,and the differences were statistically significant(t=41.355,48.614,43.587,32.561,all P<0.05);the gene expression of Bax and caspase3 in the study group were higher than those in the control group,and the gene expression of Bcl-2 was lower than that in the control group,and the differences were statistically significant(all P<0.05).The results of real-time fluorescence quantitative polymerase chain reaction showed that the expression of PI3K,Akt and mTOR were higher than those of the control group,the expression of MRPS9 was lower than that of the control group,the gene expression of Bax and caspase3 were higher than those of the control group,and the gene expression of Bcl-2 was lower than those of the control group,and the differences were statistically significant(all P<0.05).Conclusion
