径山绿茶提取物的护肤功效研究

摘  要:以径山不同区域的绿茶为原料,利用超声波细胞破壁技术得到茶叶提取物,进行化妆品抗衰老功效研究。实验结果表明,径山绿茶提取液中含有丰富的茶多酚、黄酮以及多糖成分,且不同区域的茶叶提取物中活性物含量以及比例具有一定差异。从生化水平、细胞水平以及基因水平探讨径山绿茶的抗氧化以及光老化效果。实验结果表明,径山绿茶提取液对DPPH自由基和ABTS自由基的IC_(50)约为5.0μg/mL和1.0μg/mL。在细胞水平上,径山绿茶提取物对UVB诱导的细胞损伤具有一定的保护作用,可提高光损伤后成纤维细胞的存活率。在转录水平上,与UVB诱导的模型组相比,加药组的成纤维细胞中与MMP-1和TIMP-1相关的基因出现了明显的下调和上调。以上结果表明,径山绿茶提取液具有潜在的抗氧化以及抗光老化效果。Using green tea from different regions of Jingshan as raw materials,tea extracts were obtained by using ultrasonic cell wallbreaking technology for the study of cosmetic anti-aging efficacy.The experimental results showed that the Jingshan green tea extracts were rich in tea polyphenols,flavonoids,and polysaccharides,and the contents and ratios of the actives in the tea extracts from different regions differed to some extent.The antioxidant and photoaging effects of Jingshan green tea were investigated at the biochemical,cellular,and genetic levels.The results showed that the IC_(50) of Jingshan green tea extracts were about 5.0 and 1.0μg/mL against DPPH and ABTS free radicals.At the cellular level,the Jingshan green tea extracts were protective against UVB-induced cellular damage and increased the survival rate of fibroblasts after photodamage.At the transcriptional level,genes related to MMP-1 and TIMP-1 were significantly down-and up-regulated in fibroblasts in the spiked group compared with the UVB-induced model group.The above results suggest that the Jingshan green tea extracts have potential antioxidant effects as well as anti-photo-aging effects.
