黑青稞花青素调控H9C2心肌细胞miRNA表达谱的生物信息学分析

摘  要:探讨黑青稞花青素对H9C2心肌细胞miRNA表达谱的影响,研究黑青稞花青素通过调控miRNA来实现对心肌细胞保护的潜在机制。设置6组不同处理的H9C26细胞,分别为正常组(CK)、过氧化氢组(H_(2)O_(2))、黑青稞花青素组(ACN)、原花青素组(OPC)、黑青稞花青素+过氧化氢组(ACN+H_(2)O_(2))、原花青素组+过氧化氢组(OPC+H_(2)O_(2)),处理后提取各组细胞总RNA进行miRNA测序,对测序数据进行生物信息学分析。发现H_(2)O_(2)组和CK组比较,H_(2)O_(2)处理H9C2后导致469个miRNA上调,133个miRNA下调;H_(2)O_(2)组和ACN+H_(2)O_(2)组比较,H_(2)O_(2)致273个miRNA上调,214个miRNA下调;H_(2)O_(2)组和OPC+H_(2)O_(2)组比较,H_(2)O_(2)致330个miRNA上调,222个miRNA下调;ACN和CK组比较,88个miRNA上调,25个miRNA下调;OPC和CK组比较,88个miRNA上调,27个miRNA下调;这些结果表明花青素预处理可通过调节miRNA缓解H_(2)O_(2)带来的影响。KEGG富集分析及差异表达miRNA的靶基因预测和富集分析结果显示,黑青稞花青素通过调控mTOR、PI3K/Akt/mTOR、MAPK等信号通路及上调miR-21等miRNA的表达,发挥抗氧化和抗凋亡作用。本研究可为黑青稞花青素预防和治疗心血管疾病提供一定的依据。To investigate the effect of black barley anthocyanins on the miRNA expression profile of H9C2 cardiomyocytes,and to study the potential mechanism of their protection against cardiomyocytes by regulating miRNA.Six sets of H9C26 cells were treated,namely normal group(CK),hydrogen peroxide group(H_(2)O_(2)),black barley anthocyanin group(ACN),procididin group(OPC),black barley anthocyanin+hydrogen peroxide group(ACN+H_(2)O_(2)),procidin group+hydrogen peroxide group(OPC+H_(2)O_(2)).After treatment,total RNA of each group was extracted for miRNA sequencing,and bioinformatic analysis of the sequencing data.Compared with H_(2)O_(2) and CK groups,H_(2)O_(2)Treatment of H9C2 resulted in the upregulation of 469 miRNA and 133 miRNA were downregulated;Comparison of the H_(2)O_(2) group and the ACN+H_(2)O_(2) groups,H_(2)O_(2)To 273 miRNA,214 miRNA were downregulated;Comparison of the H_(2)O_(2) group and the OPC+H_(2)O_(2) groups showed H_(2)O_(2) To 330 miRNA,222 miRNA were downregulated;Comparison of the ACN and CK groups showed 88 miRNA,25 miRNA were downregulated;Comparson of the OPC and CK groups showed 88 miRNA,27 miRNA were downregulated;These results indicated that anthocyanin pretreatment could alleviate the effects of H_(2)O_(2)by modulating miRNA.KEGG enrichment analysis and target gene prediction and enrichment analysis of differentially expressed miRNA showed that black barley anthocyanins had antioxidant and anti-apoptotic effects by regulating signaling pathways such as mTOR,PI3K/Akt/mTOR,MAPK and upregulating the expression of miRNA such as miR-21.This study provides a certain basis for the prevention and treatment of cardiovascular diseases in black highland barley anthocyanins.
