摘 要:目的:采用响应面法对以往菠萝蜜果胶提取方法进行优化,提高果胶产率以缓解果胶需求量日益增加的问题,并进一步的果胶应用提供更全面的数据分析。方法:以废弃的菠萝蜜果皮为原料,以果胶提取率为指标,在单因素的基础上,通过正交试验及响应面法建模分析各因素与果胶提取率的关系,优化超声波辅助优化菠萝蜜果皮果胶提取工艺,并与果胶红外光谱比对验证。结果:统计学结果表明菠萝蜜果皮果胶提取率与物料比、超声温度、超声时间、超声功率呈显著相关并存在交互作用,并应用Design Expert 11进行模型拟合,其模型决定系数(R2)为0.9897。结论:菠萝蜜果胶最佳提取工艺为:pH 1.0,料液比1∶25 g/mL,提取温度97℃,提取时间76.5 min,超声功率5 kHz。该条件下果胶提取率可达11.27%;菠萝蜜皮提取物具有典型的果胶红外光谱特征峰;并将其应用于制备软膏基质,有效提高了保湿效果。Objective:To optimize the extraction method of jackfruit pectin by response surface methodology,improve the yield of pectin to alleviate the increasing demand for pectin,and provide more comprehensive data analysis for further pectin application.Methods:The abandoned jackfruit peel was used as the raw material,and the pectin extraction rate was used as the index.On the basis of single factor,the relationship between each factor and pectin extraction rate was analyzed by orthogonal test and response surface method.The ultrasonic-assisted optimization of jackfruit peel pectin extraction process was optimized and compared with pectin infrared spectrum.Result:The statistical results showed that the extraction rate of pectin from jackfruit peel was significantly correlated with material ratio,ultrasonic temperature,ultrasonic time and ultrasonic power,and there was interaction.Design Expert 11 was used for model fitting,and the model determination coefficient(R2)was 0.9897.Conclusion:The optimum extraction conditions of jackfruit peel were as follows:pH 1.0,solid-liquid ratio 1∶25 g/mL,extraction temperature 97℃,ultrasonic power 5 kHz,extraction time 76.5 min.Under these conditions,the extraction rate of pectin could reach 11.27%.The obtained pectin was applied to the preparation of ointment matrix to effectively improve its moisturizing effect,and the pineapple honey peel extract extracted under the optimal conditions had typical pectin infrared light.
