摘  要:目的:运用网络药理学技术,探索了药西瓜治疗糖尿病的作用机制。方法:利用文献检索、HERB数据库和TCMSP数据库收集药西瓜的活性成分并预测其靶点,通过Gene Cards与OMIM数据库,获取糖尿病相关的靶点,通过STRING数据库构建蛋白互作网络(PPI),最后对核心靶点进行GO和KEGG富集分析。结果:获得药西瓜50种有效成分和131个作用靶点,糖尿病靶点19079个,交集靶点128个,5712条GO条目,261条KEGG信号通路,拓扑分析IL-6、CASP3、BCL2和HIF1A等靶点度值最大,活性成分主要通过干预磷脂酰肌醇-3-激酶(PI3K)/蛋白激酶B(Akt)和缺氧诱导因子-1(HIF-1)等信号通路。结论:本研究初步明确了药西瓜治疗糖尿病,筛选出了关键活性成分、核心靶点蛋白及通路,体现了中药治疗“多组分-多靶点-多通路”的作用特点,为后期开展试验验证提供了明确的方向。Objective:To explore the mechanism of Yaoxigua in treating Diabetes by using network pharmacology.Methods:Potential active ingredients and targets in Yaoxigua were screened by literature reports,TCMSP database and HERB database,the disease targets associated with Diabetes were collected by Gene Cards and OMIM databases,STRING database to construct PPI Network,and finally GO and KEGG enrichment analysis.Results:There were 50 active components and 131 action targets of Yaoxigua,and 19079 disease targets of Diabetes.The intersection targets between them were 128,GO analysis obtain 5712 items,KEGG enrichment analysis revealed 261 pathways,among them,IL-6,CASP3,BCL2 and HIF1A were the key targets.Active in gredients mainly interfere with PI3K/Akt,HIF-1 signaling pathways play a role.Conclusion:This study initially clarified the treatment of Diabetes with Yaoxigua,and screened out the key active ingredients,core target proteins and pathways,reflecting the“multi-component-multi-target-multi-pathway”feature of traditional Chinese medicine,which provided a clear direction for the verification of the experiments at a later stage.
