摘  要:目的:研究毛鸡骨草的化学成分及其体外抗炎活性。方法:综合运用硅胶、ODS、半制备高效液相等色谱方法对毛鸡骨草的甲醇提取物进行分离纯化,通过HR-ESI-MS和NMR数据分析和与文献对比确定化合物结构。采用Griess法测定化合物对脂多糖(LPS)诱导的小鼠RAW264.7巨噬细胞产生一氧化氮(NO)的抑制活性。结果:从毛鸡骨草甲醇提取物的乙酸乙酯萃取物中分离得到17个化合物,分别鉴定为:formononetin(1)、阿佛洛莫生(2)、8-羟基-4′,7-二甲氧基异黄酮(3)、8-甲雷杜辛(4)、鹰嘴豆芽素A(5)、毛蕊异黄酮(6)、7-羟基-3′,4′-二甲氧基异黄酮(7)、geraldone(8)、5,7,4′-三羟基-3′-甲氧基黄酮(9)、3-醛基吲哚(10)、methyl hematinate(11)、阿魏酸甲酯(12)、邻苯二甲酸二甲酯(13)、邻苯二甲酸二丁酯(14)、对羟基苯甲醛(15)、水杨酸(16)、对羟基苯甲酸甲酯(17)。在20μmol/L浓度时,化合物5、8、9和12对LPS诱导的RAW264.7细胞NO生成的抑制率为79.15%~88.93%。结论:其中,化合物6~9、11~13、15为首次从该属植物中分离得到,化合物5、8、9和12具有潜在的抗炎活性。Objective:To study the chemical constituents of Abrus mollis and anti-inflammatory activity.Methods:The methanol ex-tract of Abrus mollis was isolated and purified by column chromatography on silica gel,ODS and semi preparative-HPLC,The structures were identified by HR-ESI-MS and NMR spectroscopic data analyses and comparison with reported spectroscopic data.The inhibitory activity of the compounds on the production of nitric oxide(NO)in RAW264.7 macrophages of mice induced by lipopolysaccharide(LPS)was measured by Griess method.Results:Seventeen compounds were isolated from the ethyl acetate extract of the methanol ex-tract of Abrus mollis and identified as formononetin(1),afromosin(2),8-hydroxy-4′,7-dimethoxyisoflavone(3),8-methylretusin(4),biochanin A(5),calycosin(6),7-hydroxy-3′,4′-dimethoxyisoflavone(7),geraldone(8),5,7,4′-trihydroxy-3′-methoxyflavone(9),3-formylindole(10),methyl hematinate(11),methyl ferulate(12),dimethyl phthalate(13),dibutylphthalate(14),p-hydroxybenzalde-hyde(15),salicylic acid(16),methyl 4-hydroxybenzoate(17),respectively.At 20μmol/L,compounds 5,8,9 and 12 inhibited NO pro-duction in RAW 264.7 cells induced by LPS from 79.15%to 88.93%.Conclusion:Among them,compounds 6 to 9,11 to 13 and 15 are isolated from Abrus genus for the first time,compounds 5,8,9 and 12 have potential anti-inflammatory activities.
