摘 要:为了探究增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)在乾华肉用美利奴羊毛囊生长周期中的表达规律,试验以相同条件下饲养的2.5~3.5岁、体重为(68.26±4.19)kg的健康乾华肉用美利奴羊母羊为研究对象,分别于2023年9月份(兴盛期)、2024年1月份(退行期)和3月份(休止期)随机各选取3只羊,采集右侧肩胛骨后缘皮肤组织,采用免疫组织化学技术检测乾华肉用美利奴羊毛囊不同生长时期PCNA在毛囊中的定位和表达情况,并进一步采用实时荧光定量PCR和Western-blot方法分别检测毛囊不同生长时期PCNA mRNA和蛋白的表达水平。结果表明:PCNA在3个毛囊生长时期均有表达,且主要在毛母质细胞、毛干、内根鞘和外根鞘中表达;PCNA mRNA和蛋白的相对表达水平均在兴盛期最高,退行期居中,休止期最低。说明PCNA在乾华肉用美利奴羊毛囊中的表达具有规律性,且与毛囊的生长时期密切相关。In order to explore the expression patterns of proliferating cell nuclear antigen(PCNA)in the growth cycle of hair follicles of Qianhua Mutton Merino,in the experiment,2.5 to 3.5-year-old healthy ewes of Qianhua Mutton Merino with a weight of(68.26±4.19)kg were raised under the same conditions.Each of 3 sheep were randomly selected in September 2023(prosperous period),January 2024(catagen period)and March 2024(telogen period),respectively.The skin tissue from the posterior border of the right scapula was collected;immunohistochemical technique was used to detect the localization and expression of PCNA in hair follicles at different growth periods of Qianhua Mutton Merino.Furthermore,real-time PCR and Western-blot methods were used to detect the mRNA and protein expression levels of PCNA in hair follicles at different growth periods,respectively.PCNA was expressed in the three hair follicle growth periods,and was mainly expressed in hair matrix cells,hair shaft,inner root sheath and outer root sheath.The mRNA and protein expression levels of PCNA were the highest in the prosperous period,the middle in the catagen period,and the lowest in the telogen period.The results indicated that the expression of PCNA in Qianhua Mutton Merino was regular and closely related to the growth periods of hair follicles.
