摘  要:随着全球宠物犬数量增多及老龄化加剧,肿瘤发病率正逐年升高。作为人类肿瘤疾病的重要补充模型,犬肿瘤的研究受到越来越多的关注。然而,肿瘤的异质性、转移、复发和耐药等问题常导致治疗失败和预后不良,因此能展示患者肿瘤异质性的个体化肿瘤模型成为精准医疗的迫切需求。类器官是由干细胞在体外培育而成的三维培养物模型,能更准确表征原发肿瘤的结构、分子特征、基因组改变和肿瘤微环境,可用于建立生物样本库、高通量筛选药物、研究发育和发病机制及指导临床个体化用药等。目前对于现有的犬肿瘤类器官模型的研究及应用情况尚不清楚。因此,本文综述了几种已成功建立的犬肿瘤类器官及其在犬肿瘤疾病研究中的应用,为进一步探索犬肿瘤的发病机制及治疗手段提供新的参考依据。With the increasing population and aging of pet dogs worldwide,the incidence of tumors is rising year by year.As an important supplementary model for human tumor disease,research on canine tumor has attracted more and more attention.However,problems such as tumor heterogeneity,metastasis,recurrence,and drug resistance often lead to treatment failure and poor prognosis.Thus,personalized tumor models that can reflect the patients’tumor heterogeneity have become an urgent need for precision medicine.Organoids are three-dimensional(3D)culture models formed by stem cells in vitro,which can summarize the structure,molecular characteristics,genomic changes and tumor microenvironment of the primary tumors more accurately.They can be used for biobank establishment,high-throughput drug screening,development and pathogenesis research,and clinical personalized drug guidance.However,the research and application status of existing canine tumor organoid models is still unclear.Therefore,this article reviewed several successfully established canine tumor organoids as well as their applications in the study of canine tumor diseases,in order to provide new references for further exploration of the pathogenesis and treatment strategies for canine tumors.
