摘 要:为了探究雌激素受体2(ESR2)基因、丝氨酸蛋白酶抑制剂E2(SERPINE2)基因及性别决定基因-区域转录因子4(SOX4)基因多态性对努比亚山羊繁殖性能的影响,试验以402只努比亚山羊为试验对象,提取血液DNA进行混池测序,筛选ESR2基因exon1区域、SERPINE2基因exon2区域和exon3区域及SOX4基因5′UTR区域潜在的单核苷酸多态性(SNPs)位点,再通过MTA-seq测序对SNPs位点进行基因分型,并将其与繁殖性状(窝产羔数、初生重、断奶重)进行关联分析。结果表明:在ESR2基因exon1区域筛选到ESR2:g.26668749T>G和ESR2:g.26668559T>A两个SNPs位点;在SERPINE2基因exon3区域中筛选到SERPINE2:g.23652703C>T 1个SNP位点;在SOX4基因5′UTR区域筛选到SOX4:g.13838537A>G 1个SNP位点。除SOX4:g.13838537A>G外,其余位点在山羊群体中均处于Hardy-Weinberg平衡状态(P>0.05)。ESR2:g.26668749T>G位点与努比亚山羊平均窝产羔数、平均初生重、平均断奶重显著相关(P<0.05);SERPINE2:g.23652703C>T位点与努比亚山羊平均初生重显著相关(P<0.05)。说明ESR2:g.26668749T>G位点的T等位基因可能与提高平均窝产羔数和平均断奶重相关,SERPINE2:g.23652703C>T的C等位基因可能与提高羊羔平均初生重相关,这两个SNPs位点可以作为努比亚山羊繁殖性状的潜在分子标记。In order to explore the effects of genetic polymorphisms of estrogen receptor 2(ESR2)gene,serine protease inhibitor E2(SERPINE2)gene,and sex determining region Y-box transcription factor 4(SOX4)gene on the reproductive traits of Nubian goats,402 Nubian goats were selected as research objects,and blood DNA was extracted for mixed pool sequencing.Potential SNPs loci in the exon1 region of ESR2 gene,the exon2 region and exon3 region of SERPINE2 gene,and the 5′UTR region of SOX4 gene were screened.Then,the SNPs were genotypized by MTA-seq sequencing,and its association with major reproductive traits(litter size,birth weight,weaning weight)was analyzed.The results showed that two SNPs loci ESR2:g.26668749T>G and ESR2:g.26668559T>A were detected in the exon1 region of ESR2 gene.One SNP locus SERPINE2:g.23652703C>T was detected in exon3 region of SERPINE2 gene.One SNP locus SOX4:g.13838537A>G was detected in the 5′UTR region of SOX4 gene.All the sites were in Hardy-Weinberg equilibrium state(P>0.05),except SOX4:g.13838537A>G.The ESR2:g.26668749T>G locus was significantly correlated with average litter size,average birth weight and average weaning weight of Nubian goats(P<0.05).The SERPINE2:g.23652703C>T loci was significantly correlated with average birth weight of Nubian goats(P<0.05).These results indicated that the T allele of ESR2:g.26668749T>G might be associated with increasing average litter size and average weaning weight,and the C allele of SERPINE2:g.23652703C>T locus might be related to increasing average birth weight of lambs.These two SNPs sites cound be used as potential molecular markers for reproductive traits in Nubian goats.
