盐酸川芎嗪对湖羊精液冷冻保存效果的影响

摘  要:为了探究在精液冷冻稀释液中添加盐酸川芎嗪对湖羊精子冷冻保存的效果,试验采取假阴道法对6只成年湖羊分别隔天采精3次,按照稀释液中盐酸川芎嗪的添加量将精液样品进行分组,对照组不添加盐酸川芎嗪,Z1、Z2、Z3、Z4组盐酸川芎嗪的添加量分别为0.04,0.08,0.12,0.16 mmol/L,精液稀释后通过两步冷冻法保存,分别检测冷冻前湖羊精子活力,冷冻-复苏后湖羊精子活力、精子形态完整性及湖羊精子中过氧化氢酶(CAT)和超氧化物歧化酶(SOD)活性。结果表明:与对照组比较,Z1、Z2、Z3和Z4组精子活力均降低,其中Z4组精子活力显著降低(P<0.05)。各组精液样本中正常精子的占比均在92%以上,差异不显著(P>0.05)。Z3组SOD、CAT活性最高,显著高于其他组(P<0.05);Z4组SOD、CAT活性最低,其中SOD活性显著低于对照组(P<0.05)。说明在基础稀释液中添加0.12 mmol/L盐酸川芎嗪能有效提高绵羊精液冷冻保存效果。In order to explore the effects of adding chuandomazine hydrochloride to the semen cryodiluent on sperm cryopreservation in Hu sheep,in the experiment,the pseudovaginal method was used to collect semen from 6 adult Hu sheep three times every other day,and they were grouped according to the amount of chuandomazine hydrochloride in the diluent.In the control group,no chuandomazine hydrochloride was added;the additions of chuandomazine hydrochloride in groups Z1,Z2,Z3 and Z4 were 0.04,0.08,0.12,0.16 mmol/L,respectively.Semen was diluted and stored by a two-step freezing method;the sperm motility of Hu sheep before freezing,sperm motility and sperm morphological integrity of Hu sheep after thawing and resuscitation,and the activities of catalase(CAT)and superoxide dismutase(SOD)in Hu sheep sperm were detected.The results showed that compared with the control group,the sperm motility of the Z1,Z2,Z3 and Z4 groups decreased,and the sperm motility of the Z4 group was significantly decreased(P<0.05).The proportion of normal sperm in the semen samples of each group was more than 92%,and the difference was not significant(P>0.05).The activities of SOD and CAT in the Z3 group were the highest,which were significantly higher than other groups(P<0.05);the activities of SOD and CAT in the Z4 group were the lowest,and the activities of SOD were significantly lower than those in the control group(P<0.05).The results indicated that adding 0.12 mmol/L of chuandomazine hydrochloride to the basal dilution could effectively improve the cryopreservation effect of sheep semen.
