摘 要:为了初步了解我国广东省猪场中猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)的流行特点和遗传变异情况,试验于2022年2月份——2023年10月份采集广东省11个地区(广州市、肇庆市、河源市、江门市、茂名市、云浮市、韶关市、清远市、珠海市、湛江市、汕头市)27家猪场共241份疑似患猪繁殖与呼吸综合征(porcine reproductive and respiratory syndrome,PRRS)病猪的肺脏组织和血液样本并采用RT-PCR方法进行PRRSV检测,根据检测结果选取不同地区猪场的10份阳性样本病毒核酸对ORF5和Nsp2基因进行测序及序列遗传变异分析。结果表明:241份肺脏组织和血液样本的总阳性率为21.99%;不同地区阳性率在9.90%~42.86%之间,其中阳性率最高的地区为广州市,最低的地区为云浮市;肺脏组织样本阳性率为14.28%,血液样本阳性率为24.32%。从基于PRRSV ORF5基因构建的遗传进化树中可见,选取的10株PRRSV毒株中有1株(GDZH)位于以JXA1和CH-1a为代表的Lineage8.7亚群分支上,有5株(GDJM、GDKP、GDHY、GDMM、GDST)位于以NADC30为代表的Lineage1亚群分支上,有4株(GDGZ、GDGZ-2、GDZJ、GDGY)位于Lineage3亚群分支上。选取的10株PRRSV毒株ORF5基因之间的核苷酸序列相似性为80.1%~99.3%,其中GDGZ、GDZJ、GDGY和GDGZ-2的ORF5基因与Lineage3亚群毒株QYYZ(登录号为JQ308798.1)的核苷酸序列相似性最高,分别为90.2%、89.4%、93.7%、90.0%;GDJM、GDKP、GDHY、GDST和GDMM的ORF5基因与Lineage1亚群毒株NADC30(登录号为MH500776.1)的核苷酸序列相似性最高,分别为91.0%、92.2%、90.8%、85.7%、91.7%,GDZH的ORF5基因与Lineage8.7亚群高致病性毒株JXA1(登录号为EF11244.5)的核苷酸序列相似性最高,为96.2%。从基于PRRSV Nsp2基因构建的遗传进化树中可见,选取的10株PRRSV毒株中有5株(GDZH、GDGZ、GDGZ-2、GDZJ、GDGY)位于以JXA1和CH-1a为代表的Lineage8.7亚群分支上,有5株(GDJM、GDHY、GDST、GDMM、In order to preliminarily understand the epidemiological characteristics and genetic variation of Porcine reproductive and respiratory syndrome virus(PRRSV)in pig farms of Guangdong Province,China,in this study,from February 2022 to October 2023,a total of 241 lung tissue and blood samples from 27 pig farms in 11 regions of Guangdong Province(Guangzhou City,Zhaoqing City,Zhuhai City,Jiangmen City,Maoming City,Yunfu City,Shaoguan City,Qingyuan City,Zhuhai City,Zhanjiang City,Shantou City)were collected.The RT-PCR method was used for PRRSV detection.According to the test results,10 positive samples of viral nucleic acid from pig farms in different regions were selected for sequencing and sequence genetic variation analysis of ORF5 and Nsp2 genes.The results showed that the overall positive rate of 241 lung tissue and blood samples was 21.99%.The positivity rate ranged from 9.90%to 42.86%in different regions,with the highest positive rate in Guangzhou City and the lowest in Yunfu City.The positive rate of lung tissue samples was 14.28%;the positive rate of blood samples was 24.32%.From the genetic phylogenetic tree constructed based on PRRSV ORF5 gene,it could be seen that among the 10 selected PRRSV strains,1 strain(GDZH)was located on the Lineage8.7 subset clade represented by JXA1 and CH-1a;5 strains(GDJM,GDKP,GDHY,GDMM,GDST)was located on the Lineage1 subset clade represented by NADC30;and 4 strains(GDGZ,GDZJ,GDGY,GDGZ-2)was located on the Lineage3 subgroup clade.The nucleotide sequence similarity of ORF5 gene of the 10 selected PRRSV strains ranged from 80.1%to 99.3%.Among them,the ORF5 gene of GDGZ,GDZJ,GDGY and GDGZ-2 had the highest nucleotide sequence similarity with the Lineage3 subset strain QYYZ(the login number was JQ308798.1),which was 90.2%,89.4%,93.7%and 90.0%,respectively.The ORF5 gene of GDJM,GDKP,GDHY,GDST and GDMM had the highest nucleotide sequence similarity with the Lineage1 subgroup strain NADC30(the login number was MH500776.1),which was 91.0%,92.2%,90.8%,85.7%and 91.7%,respectively.The nucl
