摘 要:试验旨在研究淫羊藿总黄酮的提取方法及抗氧化活性。基于单因素试验结果,研究运用响应面分析法优化淫羊藿总黄酮的提取工艺条件,并对其抗氧化活性进行系统评价。结果表明,淫羊藿总黄酮的最佳提取工艺参数为:乙醇浓度、液料比、提取温度和时间分别是70%、25 mL/g、70℃、60 min。在此最优条件下,进行3次重复试验,测得淫羊藿总黄酮的提取得率为7.65%,与预测值7.69%非常接近。淫羊藿总黄酮提取液对2,2-二苯基-1-苦基肼基(DPPH)自由基的清除能力的IC50值为82.065 mg/L,对2,2'-联氮-双(3-乙基苯并噻唑啉-6-磺酸)(ABTS+)自由基的清除能力的IC50值为78.259 mg/L。研究表明,响应面法优化超声波辅助提取工艺方案设计可行,淫羊藿总黄酮的抗氧化活性较强,具有作为抗氧化饲料添加剂的潜力。The experiment was aimed to study the extraction method of total flavonoids from Epimedium and its antioxidant activity.Based on the results of single factor experiment,response surface analysis was used to optimize the extraction conditions of total flavonoids of Epimedium,and its antioxidant activity was systematically evaluated.The results showed that the optimal extraction parameters were ethanol concentration,liquid-solid ratio,extraction temperature and extraction time of 70%,25 mL/g,70℃and 60 min,respectively.Under the optimal conditions,the extraction rate of total flavonoids of Epimedium was 7.65%,which was very close to the predicted value of 7.69%.The IC50 value of the extract of total flavonoids from Epimedium for scavenging 2,2-diphenyl-1-picrohydrazyl(DPPH)free radicals was 82.065 mg/L,and the IC50 value for scavenging 2,2'-diazo-bis(3-ethylbenzothiazolin-6-sulfonic acid)(ABTS+)free radicals was 78.259 mg/L.The study indicates that the optimization of the ultrasound-assisted extraction process by response surface methodology is feasible,and total flavonoids from Epimedium had strong antioxidant activity and the potential to be used as an antioxidant feed additive.
