摘 要:试验旨在筛选出促进盐碱胁迫下紫花苜蓿生长及生理效应的最适宜褪黑素(MT)浸种浓度。以紫花苜蓿三得利和甘农三号为试验材料,采用0(T1组)、50(T2组)、100(T3组)、150(T4组)、200μmol/L(T5组)的MT溶液进行浸种处理,并对幼苗进行盐碱胁迫,CK组仅采用蒸馏水浸种未施加盐碱胁迫,每组3个重复,测定盐碱胁迫下紫花苜蓿幼苗的根长、株高及丙二醛(MDA)、可溶性糖、脯氨酸(Pro)含量等生理指标。结果表明:T4组紫花苜蓿幼苗根长、株高、干重和鲜重显著低于CK组(P<0.05),显著高于T1组(P<0.05)。T4组的MDA含量相比于T1组显著降低(P<0.05),而超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶活性(CAT)活性显著高于T1组(P<0.05)。T3组的紫花苜蓿幼苗的可溶性糖和Pro含量显著高于CK组(P<0.05)。隶属函数评价结果显示,三得利和甘农三号的耐盐碱性排序均为CK组>T4组>T3组>T2组>T5组>T1组,表明150μmol/L MT溶液浸种处理对盐碱胁迫的缓解效果较好。研究表明,甘农三号对环境变化的反应更敏感,表现出较强的盐碱胁迫响应,而三得利则表现出更强的抗性。The experiment aims to screen for the most suitable melatonin(MT)soaking concentration that promotes the growth and physiological effects of Medicago sativa under salt-alkali stress.Using Medicago sativa Suntory and Gannong No.3 as experimental materials,seeds were soaked with MT solution of 0(T1 group),50(T2 group),100(T3 group),150(T4 group),200μmol/L(T5 group),and seedlings were subjected to salt-alkali stress,while seeds were soaked with distilled water only without salt-alkali stress in CK group,with three replicates per group.The physiological indicators such as root length,plant height,and malondialdehyde(MDA),soluble sugart,and proline(Pro)content of Medicago sativa seedlings under alt-alkali stress were measured.The results showed that the root length,plant height,dry weight,and fresh weight of Medicago sativa seedlings in T4 group were significantly lower than those in CK group(P<0.05),but significantly higher than those in T1 group(P<0.05).The MDA content in T4 group was significantly decreased compared with that in T1 group(P<0.05),while the activities of superoxide dismutase(SOD),peroxidase(POD)and catalase activity(CAT)in T4 group were significantly higher than those in T1 group(P<0.05).The concentrations of soluble sugar and Pro in alfalfa seedlings in T3 group were significantly higher than those in CK group(P<0.05).The results of membership function evaluation showed that the salt-alkali tolerance of Suntory and Gannong No.3 was ranked as CK group>T4 group>T3 group>T2 group>T5 group>T1 group,indicating that 150μmol/L MT solution had the best effect on saltalkali stress relief.The study indicates that Gannong No.3 is more sensitive to environmental changes,showing a stronger response to saline-alkali stress,while Suntory showed a stronger resistance.
