摘  要:为了解小麦生长素外输基因PIN1的基本结构和生物学功能。本研究以‘贵紫麦1号’的cDNA序列全长为模板,通过RT-PCR方法得到GzPIN1-7B基因,利用生物信息学分析软件对该基因及其编码的蛋白质进行分析。结果表明:GzPIN1-7B蛋白共编码586个氨基酸,属于疏水性蛋白,亚细胞定位在细胞膜上;该蛋白不存在信号肽,是一种分泌蛋白;蛋白功能结构域分析显示其含有两个Mem-trans结构域,属于Mem-trans超家族;二、三级结构预测显示主要由无规卷曲和α-螺旋组成;其编码的氨基酸序列与大麦、小麦、节节麦的同源关系较近;其启动子顺式作用元件分析结果显示,GzPIN1-7B启动子中除了含有CAAT-box和TATA-box等核心启动子元件外,还有响应激素和逆境胁迫等相关的顺式作用元件。以上研究结果为进一步探索GzPIN1-7B基因参与小麦生长发育的调控机制提供理论依据。In order to study the basic structure and biological functions of wheat auxin transmission gene PIN1.In this study,GzPIN1-7B gene was obtained by RT-PCR with the full-length cDNA sequence of'Guizimai No.1'as a template,and the gene and its encoded protein was analyzed by using bioinformatics analysis software.The results showed that:GzPIN1-7B protein encoding 586 amino acids,belonging to the hydrophobic protein,sub-cellular localization in the cell membrane,which is a kind of secretory proteins without signal peptide.Function structure domain analysis showed that it belonged to the Mem-trans super family,and contains two conserved Mem-trans domains.Secondary and tertiary structures analysis showed it was mainly composed of random coil and alpha helix.The homologous of amino acid was close to Hordeum vulgare L.and Triticum aestivum L.and A egilops tauschii subsp..Promoter cis-element analysis indicated that GzPIN1-7B not only contains CAAT box and TATA box promoter elements,but also contains hormone and adversity stress cis element.The above results can provide theoretical basis for further explore GzPIN1-7B gene participate in the regulatory mechanism of wheat growth and development.
