摘 要:去氢骆驼蓬碱(harmine)存在于多种药用植物中,其作用于人肝癌细胞的研究尚不清楚.用不同浓度的去氢骆驼蓬碱作用于肝癌细胞株HepG2,24 h后用MTT法检测细胞活力,用流式细胞仪检测细胞凋亡,用蛋白质印迹(western blotting,WB)的方法检测FAK/AKT和ERK1/2/CREB磷酸化水平.用伤口愈合法测定肝癌细胞的迁移,最后用RT-qPCR检测MMP2、MMP9和VEGF mRNA表达水平.结果显示:harmine能抑制肝癌HepG2细胞生长,并且呈现剂量依赖.去氢骆驼蓬碱可以通过抑制FAK/AKT和ERK1/2/CREB磷酸化水平对人肝癌细胞增殖表现出显著的抑制作用.此外,去氢骆驼蓬碱可以抑制人肝癌细胞的迁移以及抑制MMP-2、MMP-9和VEGF的表达,诱导细胞凋亡.总之,本研究结果表明,去氢骆驼蓬碱对人肝癌细胞的生长发育具有显著抑制作用.Harmine is present in various medicinal plants,and its effects on human liver cancer cells remain unclear.Different concentrations of harmine were treated to HepG2 cells for 24 h.Cell viability was measured by MTT assay and apoptosis was detected by flow cytometry.FAK/AKT and ERKv/CREB phosphorylation expression levels were detected by Western blotting and human liver cancer cell migration was assayed using a wound healing test.Finally,RT-qPCR was used to detect the mRNA expression levels of MMP2,MMP9,and VEGF.The results showed that harmine exhibited significant inhibitory effects on the proliferation of human liver cancer cells by inhibiting the phosphorylation levels of the FAK/AKT and ERKiv/2/CREB.Furthermore,harmine also inhibited the migration of human liver cancer cells and suppressed the expression levels of MMP-2,MMP-9,and VEGF.In conclusion,our findings suggest that harmine exerts a significant inhibitory effect on the development of human liver cancer cells.
