摘  要:以‘圣诞红’草莓叶片和茎尖为试材,采用离体培养技术,研究了外源激素对愈伤组织诱导、不定芽分化、增殖和生根培养的影响,建立‘圣诞红’草莓离体再生体系和组培快繁体系,以期为‘圣诞红’草莓良种资源快速繁殖和遗传转化提供参考依据。结果表明:叶片不定芽诱导阶段,1.5 mg·L^(-1) TDZ和0.1 mg·L^(-1) IBA组合最适宜愈伤组织诱导和不定芽分化;不定芽增殖阶段,最适培养基为MS+1.0 mg·L^(-1) TDZ+0.1 mg·L^(-1) IBA;不定芽生根阶段,最适培养基为MS+0.1 mg·L^(-1) IBA。茎尖最适初代培养基为MS+1.0 mg·L^(-1) 6-BA+0.1 mg·L^(-1) IBA,诱芽率为80%;获得丛生芽的最适增殖培养基为MS+0.5 mg·L^(-1) 6-BA+0.1 mg·L^(-1) IBA,增殖系数为4.20;最适生根培养基为1/2MS,生根率高达100%。Taking‘Poinsettia’strawberry leaves and stem tips as test materials,the effects of exogenous hormones on healing tissue induction,adventitious shoot differentiation,proliferation and rooting culture were studied by using the in vitro culture technique,so as to establish an in vitro regeneration system and a rapid multiplication system for‘Poinsettia’strawberry,in order to provide reference for the rapid propagation and genetic transformation of‘Poinsettia’strawberry seed resources.The results showed that the combination of 1.5 mg·L^(-1) TDZ and 0.1 mg·L^(-1) IBA was the most suitable for callus induction and indeterminate shoot differentiation in the indeterminate shoot induction stage;the most suitable medium for the indeterminate shoot proliferation stage was MS+1.0 mg·L^(-1) TDZ+0.1 mg·L^(-1) IBA;in the indeterminate shoot rooting stage,the most suitable medium for the rooting medium was MS+0.1 mg·L^(-1) IBA.The most suitable medium for the primary culture of stem tip was MS+1.0 mg·L^(-1)6-BA+0.1 mg·L^(-1) IBA,with a germination rate of 80%;the optimum proliferation medium for obtaining clumped shoots was MS+0.5 mg·L^(-1)6-BA+0.1 mg·L^(-1) IBA,with a proliferation coefficient of 4.20;and the optimum rooting medium was 1/2MS,with a rooting rate as high as 100%.
