麒麟针药对POI模型大鼠卵巢功能的影响及机制研究

摘　　要：目的观察麒麟针药对环磷酰胺诱导的早发性卵巢功能不全(POI)模型大鼠的卵巢功能影响及可能机制。方法55只POI造模成功的雌性Wistar大鼠随机分为中药组、针刺组、针药组、西药组、模型组及正常组,每组各11只,分别给予中药、针刺、针刺加中药、芬吗通、0.9%氯化钠溶液处理,连续4周。HE染色观察组织形态学变化,ELISA检测血清中FSH、LH、E 2水平含量,蛋白免疫印迹法(Western blot)检测卵巢PI3K、Akt、mTOR、HIF-1α、VEGF蛋白表达水平。结果与正常组比较,模型组大鼠动情周期紊乱,卵巢体积减小,各级卵泡数明显减少,血清FSH、LH水平升高,E 2降低,卵巢中PI3K、Akt、mTOR、HIF-1α、VEGF蛋白表达显著降低(P<0.01);与模型组比较,中药组、针刺组、针药组卵泡增多,血清E 2升高,FSH、LH降低,PI3K、Akt、mTOR、HIF-1α、VEGF蛋白表达升高(P<0.01)。结论麒麟针药可调节POI大鼠血清性激素水平,明显改善卵巢功能,其机制可能与PI3K/Akt/mTOR/HIF-1α/VEGF通路有关。Objective To observe the effect of Qilin Decoction(麒麟方)combined with Qilin acupuncture on ovarian function of rats with premature ovarian insufficiency(POI)induced by cyclophosphamide and its possible mechanism.Methods 55 POI model rats were randomly divided into the Chinese medicine group(Qilin Decoction),the acupuncture group(Qilin acupuncture),the decoction combined with acupuncture group(Qilin Decoction and Qilin acupuncture),the western medicine group(Femoston),and the model group(normal saline),another 11 healthy rats were classified into the normal group(normal saline).The rats in each group were given the corresponding treatment for 28 successive days.The ovarian histomorphological changes were determined by HE staining,the levels of follicle stimulating hormone(FSH),luteinizing hormone(LH)and estradiol(E 2)in serum were detected by enzyme linked immunosorbent assay(ELISA),and the protein expression levels of phosphatidylinositol-3-kinase(PI3K),protein kinase B(Akt),mammalian target of rapamycin(mTOR),hypoxia inducible factor-1α(HIF-1α)and vascular endothelial growth factor(VEGF)in the rat ovarian tissue were determined by Western blot.Results Compared with those of the normal group,the estrous cycle of rats in the model group was disordered,the ovarian volume was reduced,the number of follicles was significantly reduced,the levels of serum FSH and LH were significantly increased,the level of E 2 was decreased,and the expressions of PI3K,Akt and mTOR protein in the rat ovarian tissue were significantly decreased(P<0.01).Compared with those of the model group,the number of follicles in the Chinese medicine group,the acupuncture group and the decoction combined with acupuncture group were significantly increased,the levels of serum FSH and LH were significantly down-regulated,the levels of serum E 2 were significantly up-regulated,and the expressions of PI3K,Akt,mTOR,HIF-1αand VEGF proteins were significantly increased too(P<0.01).Conclusion Qilin Decoction combined with Qilin acupuncture can signi
