摘 要:该研究从山东省威海市麻子港海岸带海洋沉积物中采样,筛选得到高产蛋白酶菌株,并对其特性、生长条件以及酶活力进行探究。通过形态学观察、生理生化试验、16S rDNA序列分析和基因组分析鉴定菌株,采用单因素结合响应面实验优化菌株的最佳生长条件,并于此条件下测定该菌株的蛋白酶活力。该菌株为高地芽孢杆菌(Bacillus altitudinis),盐度生长范围为0~160 g/L,最适生长条件为:温度40℃,培养基初始pH值9,摇床转速229 r/min,盐度为30 g/L,最适碳源为葡萄糖,最适氮源为蛋白胨,在该条件下蛋白酶的酶活力可达99.747 U/mL。筛选获得的高地芽孢杆菌A1具备良好的产蛋白酶的能力和耐盐耐热性,可以为餐厨垃圾降解、工业废水处理、微生物菌肥制作以及生产工艺调整提供生物学依据。Marine sediments were sampled from the coastal of Mazigang,Weihai city,Shandong province.This study aimed to screen proteinase-producing bacterial strains and explore their characteristics,growth conditions,and enzyme activities.The strains were identified through morphological observation,physiological and biochemical tests,16S rDNA sequence analysis,and genomic analysis.The optimal growth conditions of the strain were optimized by using single factor combined with response surface experiment,and the protease activity of the strain was measured under these conditions.The isolated strain was identified as Bacillus altitudinis,with a salinity growth range of 0-160 g/L.The optimal growth conditions for this strain were as follows:temperature 40℃,initial pH 9,shaker speed 229 r/min,salinity 30 g/L,optimal carbon source for glucose,and peptone as the preferred nitrogen source.Under these conditions,the protease activity reached 99.747 U.The screened Bacillus altitudinis A1 had good protease production ability,salt tolerance,and heat resistance,which could provide biological basis for food waste degradation,industrial wastewater treatment,microbial fertilizer production,and production process adjustment.
