摘  要:本文对濒危兰科植物的根、茎、叶、花、种子不同组织的DNA提取效果进行了研究,为开展分子鉴定等研究提供基础。采用改良十六烷基三甲基溴化铵法(Cetyltrimethylammonium Bromide,CTAB)提取其基因组DNA,对获得的DNA质量、浓度和纯度进行比较;并以获得的基因组DNA作为模板进行DNA条形码基因ITS、matK、psbA-trnH的PCR扩增验证,评价各基因序列的扩增效率。研究结果表明,不同组织提取的DNA质量均较好,DNA电泳条带清晰、明亮。种子提取的DNA浓度最高,根、叶、茎次之,花最低。不同组织提取基因组DNA模板的PCR扩增效果良好,可用于鉴定濒危兰科植物物种的DNA条形码扩增等实验。To establish a basis for molecular identification and associated studies,the research examines the efficacy of DNA extraction from diverse tissues-roots,stems,leaves,flowers,and seeds of endangered orchid species.The modified CTAB protocol was utilized for genomic DNA extraction,and a comparative analysis was conducted regarding the quality,concentration of the extracted DNA.The isolated genomic DNA was employed as a template for PCR amplification of DNA barcode genes,including ITS,matK,and psbA-trnH,to assess the amplification efficiency of each gene segment.The findings demonstrate that the quality of DNA extracted from different tissues,post-optimization,is predominantly high,evidenced by distinct and prominent DNA electrophoresis bands.The highest DNA concentration was recorded from seeds,followed by roots and leaves,with flowers yielding the lowest concentration.The PCR amplification of genomic DNA extracted from different tissues showed good performance,rendering it suitable for experiments focused on DNA barcode amplification for the identification of endangered orchid species.
