摘  要:为明确野外新采集夜蛾黑卵蜂Telenomus remus孤雌产雌品系沃尔巴克氏体Wolbachia感染情况及其对生殖模式的影响,首先利用沃尔巴克氏体表面蛋白(Wolbachia surface protein,wsp)基因以及细胞分裂蛋白基因ftsz特异性引物对该品系DNA进行PCR扩增,并基于单基因和多位点序列分型法对沃尔巴克氏体类群进行系统发育分析,并通过高温连续处理夜蛾黑卵蜂2个世代,研究其对夜蛾黑卵蜂性比的影响。结果表明:夜蛾黑卵蜂孤雌产雌品系体内存在沃尔巴克氏体感染,该品系的wsp序列与纳氏黑卵蜂Telenomus nawai体内沃尔巴克氏体株系wNaw达到100%的相似度。2种系统发育树结果均显示夜蛾黑卵蜂感染的沃尔巴克氏体株系属于A超组,与ftsz基因特异性引物检测结果一致。高温处理后,夜蛾黑卵蜂后代雄性比例逐代升高,F_(1)代产雄率为9.23%,F_(2)代产雄率为94.45%,且出现了少量雌雄嵌合体个体。表明新发现的夜蛾黑卵蜂孤雌产雌品系可作为害虫生物防治的新的天敌资源。To determine the Wolbachia infection status of a newly collected thelytokous strain of egg parasitoid Telenomus remus and its influence on reproductive mode,we conducted polymerase chain re‐action(PCR)amplification using specific primers targeting the Wolbachia surface protein(wsp)gene and the cell division protein gene ftsz.Phylogenetic analysis was performed based on both single-gene and multilocus sequence typing method.Additionally,we investigated the effect of high-temperature on the sex ratio of T.remus.The results confirmed Wolbachia infection in the thelytokous strain of T.re‐mus,with the wsp sequence exhibiting 100%similarity to that of strain wNaw from T.nawa.Phyloge‐netic tree analysis consistently placed the Wolbachia strain infecting T.remus within supergroup A,in agreement with the ftsz gene-specific primerdetection results.Following high-temperature treatment,the proportion of male offspring increased progressively across generations, with male emergence rates of9.23% in the F_(1) generation and 94.45% in the F_(2) generation. A small number of intersex individuals werealso observed. This study represents the first report of a thelytokous strain of T. remus collected from thefield in China. The discovery of this strain provides a novel natural enemy resource for the biologicalcontrol of agricultural pests.
