摘 要:从人参中分离得到一种新的活性多糖WGP60,并对其比旋光度、分子量分布、单糖组成和寡糖片段等结构特征进行分析,以成纤维细胞NIH3T3对人参多糖WGP60的药理活性进行研究.人参多糖WGP60由葡萄糖、半乳糖和阿拉伯糖组成,物质的量比为77.1∶16.8∶6.1,比旋光度为+60.354°,重均分子量约为79108 g/mol.多糖结构中存在Gal-Gal、Ara-Gal-Ara、Gal-Gal-Glc-Ara等结构片段.药理实验结果表明,人参多糖WGP60在1.6×10^(-2)g/L的质量浓度下可显著促进成纤维细胞NIH3T3的增殖;在2.0×10^(-3)g/L的质量浓度下,显著提高紫外线(UVA)损伤的成纤维细胞NIH3T3存活率.研究表明,人参多糖WGP60是一种新的人参多糖成分,对成纤维细胞NIH3T3具有良好的促进增殖和修复UVA损伤作用,为人参临床应用及进一步研究开发提供科学依据.A new active polysaccharide WGP60 was isolated from Panax ginseng,and its specific rotation,molecular weight distribution,monosaccharide composition and oligosaccharide fragment were analyzed to study the pharmacological activity of WGP60 by fibroblasts NIH3T3.Ginseng polysaccharide WGP60 is composed of glucose,galactose and arabinose.Their molar ratio is 77.1∶16.8∶6.1,the specific rotation is+60.354°,and the weight average molecular weight is 79108 g/mol.There are Gal-Gal,Ara-Gal-Ara,Gal-Gal-Glc-Ara and other structural fragments in the polysaccharide structure.The results of pharmacological experiments showed that ginseng polysaccharide WGP60 could significantly promote the proliferation of fibroblasts NIH3T3 at the mass concentration of 1.6×10^(-2)g/L;at the mass concentration of 2.0×10^(-3)g/L,the survival rate of fibroblasts NIH3T3 damaged by ultraviolet A(UVA)was significantly improved.Ginseng polysaccharide WGP60 is a new component of ginseng polysaccharide,which has good effects on promoting proliferation and resisting UVA damage of fibroblasts NIH3T3.Our study provides scientific basis for clinical application and further research and development of Panax ginseng.
