摘　　要：大鼠肉瘤基因(Rat sarcoma,RAS)是人类癌症中最常见的突变致癌基因,也是著名的“不可成药”靶点.Lumakras是一种新型的鼠类肉瘤病毒癌基因(Kirsten rat sarcoma viral oncogene homolog,KRAS)抑制剂,2021年,美国食品药品监督管理局(FDA)批准了安进公司研制的抗肿瘤药物Lumakras(Sotorasib)上市,成为全球第一个治疗携带KRAS G12C突变的中晚期非小细胞肺癌患者的靶向药物.本文就Lumakras的合成路线、临床研究、不良反应和用药注意事项等方面作简要介绍,希望为其今后的临床应用提供参考.Rat sarccma(RAS)is the most common mutated oncogene in human cancer,and is also a well-known“undruggable”target.Lumakras is a novel Kirsten rat sarcoma viral oncogene homolog(KRAS)inhibitor.In 2021,the U.S.Food and Drug Administration(FDA)has approved Amgen’s antitumor drug Lumakras(Sotorasib),becoming the world’s first targeted drug for the treatment of patients with intermediate and advanced non-small cell lung cancer with KRAS G12C mutations.Therefore,this article briefly introduces the synthesis route,clinical research,adverse reactions,medication precautions and other aspects of Lumakras,providing reference for its clinical application.
