摘 要:建立了对中药蛇床子配方颗粒中花椒毒素、佛手柑内酯和蛇床子素的高效液相色谱分析方法。使用Welch Ultimate Plus C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-水进行梯度洗脱,流速为1.2 mL/min,检测波长为248 nm,柱温30℃,进样体积12μL。通过改进流动相和梯度洗脱程序,用普通粒径C18柱在较短的时间(30 min)内,可使中药蛇床子配方颗粒中花椒毒素、佛手柑内酯和蛇床子素3种成分,与相邻峰均能达到基线分离。方法学评价结果表明,建立的方法准确可靠,实验条件简便,并用于中药蛇床子配方颗粒实际样品的分析。A high-performance liquid chromatography(HPLC)analytical method was established for the determination of xanthotoxin,bergapten,and osthole in Cnidii Fructus formula granule,a traditional Chinese medicine.The HPLC determination was performed using Welch Ultimate Plus C18 column(250 mm×4.6 mm,5μm).The mobile phase consisted of acetonitrile-water and gradient elution.The flow rate was set at 1.2 mL/min,the detection wavelength was 248 nm,the column temperature was maintained at 30℃,and the injection volume was 12μL.By improving the mobile phase composition and the gradient elution program,with the use of a common particle size C18 column,the three components,namely xanthotoxin,bergapten,and osthole in Cnidii Fructus formula granule,could achieve baseline separation from the adjacent peaks within a relatively short time(less than 30 min).The results of the methodological evaluation indicated that the established method was accurate and reliable,and the experimental conditions were simple and convenient.This method was also applied to the analysis of actual samples of the formula granules of Cnidii Fructus.
