福建柏不定芽生根诱导及其内源激素研究

摘  要:为建立福建柏不定芽诱导生根过程中最适激素配比及探究诱导生根过程中外源激素对内源激素的影响,取福建柏上当年生幼嫩无害的带芽茎段进行组织培养,以所获得的不定芽作为研究对象,测定不同配方下不定芽的生根率、污染率及内源激素的变化情况。结果表明:单独添加1.0 mg·L^(-1)IBA、添加0.5mg·L^(-1)IBA+0.1 mg·L^(-1)NAA或者0.5 mg·L^(-1)IBA+0.1 mg·L^(-1)IAA能诱导不定芽生根;最佳生长素组合为1.0 mg·L^(-1)IBA+0.1 mg·L^(-1)NAA;最佳生长调节剂组合为0.1 mg·L^(-1)6-BA+0.5 mg·L^(-1)IBA;生根粉对不定芽诱导生根无明显作用;最佳诱导生根活性炭浓度为0.2 g/L;最佳诱导生根蔗糖浓度为30 g/L;诱导生根过程中,GA3、IAA、ABA激素含量有明显变化,CTK含量变化较小。研究初步建立了福建柏组培苗最佳生根激素配比,探究了内源激素变化,可为工厂化育苗提供技术支持。In order to determine the optimal hormone ratios and the effects of exogenous and endogenous hormones during the induced rooting of adventitious shoots of Fokienia hodginsii.The adventitious shoots were obtained from young and healthy stem segments of the current year sourced from Fujian cypress plants and cultured in tissue culture.The rooting rate of adventitious shoots,the contamination rate,and the changes of endogenous hormones in the plants were observed and measured under different conditioning formulas.The results showed that supplementation of 1.0 mg·L^(-1)IBA alone,0.5 mg·L^(-1)IBA+0.1 mg·L^(-1)NAA,or 0.5 mg·L^(-1)IBA+0.1 mg·L^(-1)IAA in combination could induce rooting of adventitic buds.The optimal auxin combination was 1.0 mg·L^(-1)IBA+0.1 mg·L^(-1)NAA,while the optimal plant growth regulator combination was 0.1 mg·L^(-1)6-BA+0.5 mg·L^(-1)IBA.The root powder had no obvious effect on the induced rooting of adventitious buds,and direct dipping in root powder increased the contamination rate.The optimal concentration of activated carbon was 0.2g,and the optimal rooting sucrose concentration was 30g/L.During the induction of rooting,the contents of GA3,IAA and ABA were significantly altered,while the contents of cytokinins(CTK)changed little.This study has preliminarily determined the optimal hormone ratios for the rooting of F.hodginsii tissue-culture seedlings,investigated the changes in endogenous hormones,and can thus offer technical support for industrialized nursery cultivation.
