摘  要:以滇黄精(Polygonatum kingianum)原球茎为材料,采用薄层细胞培养技术,研究不同薄层位置与不同植物生长调节剂对薄层不定芽诱导的影响;采用流式细胞仪法和染色体计数法,对滇黄精再生植株进行倍性稳定检测。结果表明:原球茎顶端切取的第1片薄层部位出芽能力最强,在含有1.0 mg·L^(−1)吲哚乙酸(IBA)+1.0 mg·L^(−1)6-苄基腺嘌呤(6-BA)的不定芽诱导培养基中平均出芽数最多,达到6.69个。不定芽在1/2MS+1.0 mg·L^(−1)IBA+1 g·L^(−1)活性炭的培养基中长至完整植株,再生植株经驯化后移栽至室外,成活率达90%。薄层细胞培养技术获取的再生植株,与母本植株的倍性保持一致。本研究建立了滇黄精再生植株体系,能够进行黄精种苗的规模化生产,并为滇黄精的基因育种工程提供技术参考。The study investigated the effects of various thin-layer positions and plant growth regulators on the induction of thin-layer adventitious shoots using thin cell layer culture technology with Polygonatum kingianum protocorms.The ploidy stability of the regenerated P.kingianum was assessed using flow cytometric analysis and chromosome counting methods.Results revealed that the first lamina cut from the protocorm tip exhibited the highest germination ability,with the greatest average number of adventitious shoots observed in the medium containing 1.0 mg·L^(−1)indoleacetic acid(IBA)+1.0 mg·L^(−1)6-benzyl adenine(6-BA),reaching 6.69 shoots.Adventitious shoots developed into complete plants in a medium of 1/2MS+1.0 mg·L^(−1)IBA+1 g·L^(−1)activated charcoal,achieving a 90%survival rate upon outdoor transplantation.The regenerated plants obtained through the thin cell layer culture technology maintained the same ploidy as the parent plants.This study established a regeneration system for P.kingianum which facilitates the large-scale production of seedlings and provides a technical reference for the genetic breeding engineering of P.kingianum.
