摘 要:目的研究山奈酚对脂多糖(LPS)诱导小鼠肝损伤的保护作用,为开发新型保肝药物提供实验和理论依据。方法将小鼠分为正常组、模型组、阳性对照组和山奈酚低、中、高剂量组,每组10只。治疗15 d,测定血清碱性磷酸酶(ALP)、天门冬氨酸氨基转移酶(AST)和丙氨酸氨基转移酶(ALT)活性,肝脏总抗氧化(T-AOC)、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)的水平,Toll样受体4(TLR4)、核因子κB(NF-κB)和肿瘤坏死因子α(TNF-α)mRNA和蛋白表达;观察肝脏形态学变化。结果与正常组比较,模型组小鼠ALT、AST和ALP活性显著增加(t=29.075、26.676、21.349),T-AOC、SOD和CAT水平显著下降(t=13.851、25.765、18.444),TLR4、NF-κB和TNF-α的mRNA(t=12.200、23.003、13.244)和蛋白(t=13.239、64.987、23.219)表达显著上升,差异均有统计学意义(P均<0.05)。与模型组比较,山奈酚中、高剂量组AST、ALT和ALP活性降低,T-AOC、SOD和CAT水平升高,TLR4、NF-κB和TNF-α的mRNA和蛋白表达下降,差异均有统计学意义(P均<0.05)。结论山奈酚能提高机体抗氧化能力、抑制TLR4/NF-κB通路,进而改善LPS诱导的小鼠肝损伤。Objective To study the protective effect of kaempferol on lipopolysaccharide(LPS)-induced liver injury in mice,provide an experimental and theoretical basis for the development of new liver-protection drugs.Methods The mice were divided into normal group,model group,positive group and kaempferol low,medium and high dose groups,ten individuals in each group.After 15 days of treatment,the activities of serum alkaline phosphatase(ALP),aspartate aminotransferase(AST)and alanine aminotransferase(ALT),liver total antioxidant capacity(T-AOC),superoxide dismutase(SOD)and catalase(CAT)levels,Toll-like receptor 4(TLR4),nuclear factor kappa B(NF-κB)and tumor necrosis factor-α(TNF-α)mRNA and protein expression were measured.Liver morphology were analyzed.Results Compared with the normal group,ALT,AST and ALP of mice in the model group increased significantly(t=29.075,26.676,21.349),and T-AOC,SOD and CAT decreased significantly(t=13.851,25.765,18.444).The expression of TLR4,NF-κB and TNF-α mRNA(t=12.200,23.003,13.244)and protein(t=13.239,64.987,23.219)increased significantly(all P<0.05).Compared with the model group,the activities of AST,ALT and ALP in the mid-and high-dose kaempferol groups were reduced,the levels of T-AOC,SOD and CAT were increased,and the mRNA and protein expressions of TLR4,NF-κB and TNF-α were decreased significantly(all P<0.05).Conclusion Kaempferol could improve the body's antioxidant capacity,inhibit the TLR4/NF-κB pathway,and thereby improve LPS-induced liver damage in mice.
