摘 要:目的基于网络药理学研究紫苏醛(PAE)对人宫颈癌HeLa细胞的抑制作用及其对凋亡蛋白的影响。方法利用网络药理学及分子对接探究PAE潜在的靶点蛋白;采用CCK-8法和流式细胞仪来评估PAE对HeLa细胞增殖的抑制效果,并通过JC-1荧光探针、Hoechst 33342荧光染色法和蛋白免疫印迹(Western blot)等测定HeLa细胞的凋亡情况。结果网络药理学分析和分子对接结果表明,潜在的作用靶点有丝裂原活化蛋白激酶3(MAPK3)、雄激素受体(AR)、孕激素受体(PGR)等;PAE能够抑制细胞生长,阻滞细胞周期在G0/G1期降低线粒体膜电位并诱导细胞凋亡。结论PAE可能是通过抑制线粒体途径诱导HeLa细胞凋亡。Objective To determine the inhibitory effect of perillaldehyde(PAE)in human cervical cancer HeLa cells,and on apoptotic proteins.Methods The potential target proteins were determined by network pharmacology and molecular docking.The inhibitory effect of PAE on the proliferation was determined by CCK-8 assay.The effect of PAE on the cycle distribution was detected by flow cytometry.The apoptosis of HeLa cells was determined by JC-1 fluorescent probe,Hoechst 33342 fluorescent staining method,and Western blot.Results Network pharmacology and molecular docking showed that the potential targets included MAPK3(mitogen-activated protein kinase 3),AR(androgen receptor),PGR(progesterone receptor).PAE inhibited the cell proliferation,blocked the cell cycle at the G0/G1 phase,decreased the mitochondrial membrane potential and induced apoptosis in HeLa cells.Conclusion PAE may induce the apoptosis in HeLa cells by inhibiting the mitochondrial pathway.
