青翘清热解毒的谱效关系研究

摘  要:目的研究青翘指纹图谱与清热解毒之间的谱效关系。方法采用HPLC法建立青翘的指纹图谱;再用综合指数法计算青翘清热解毒的总效应值;最后用灰色关联分析法将青翘指纹图谱中各共有峰与其清热解毒总效应值进行关联分析。结果青翘指纹图谱有26个共有峰,12批青翘相似度均在0.9以上;青翘低、中、高剂量组的抗炎总效应值分别为3.39、4.25、5.18;解热总效应值分别为1.51、0.99、0.83;17(连翘酯苷A)、1、26(连翘脂素)、22(连翘苷)号共有峰与解热药效关系的关联度大于0.8;1、22(连翘苷)、15(芦丁)号共有峰,与抗炎药效的关联度大于0.8。结论1、15(芦丁)、17(连翘酯苷A)、22(连翘苷)、26(连翘脂素)号共有峰与青翘清热解毒的疗效密切相关,可能为青翘清热解毒的物质基础。Objective To determine the spectrum-effect relationship between the chemical fingerprint of Green Forsythia decoction and its heat-clearance and detoxification effect.Methods The chemical fingerprint of Green Forsythia was established by high-performance liquid chromatography(HPLC).The total effect values of Green Forsythia’s heat-clearance and detoxification effect were calculated with a comprehensive index method.Subsequently,grey correlation analysis was performed to evaluate the relationship between the common peaks in the fingerprint and total effect values.Results Totally 26 common peaks were identified in the fingerprint of Green Forsythia,with a similarity exceeding 0.9 across 12 batches.The total anti-inflammatory effect values for low,medium,and high doses of Green Forsythia were 3.39,4.25,and 5.18,respectively.The corresponding total pyretolysis effect values were 1.51,0.99,and 0.83.Peaks 17(forsythiaside A),1,26(forsythiaside),and 22(forsythin)showed correlation coefficients greater than 0.8 with antipyretic effect.Similarly,peaks 1,22(phillyrin),and 15(rutin)showed correlation coefficients exceeding 0.8 with anti-inflammation effect.Conclusion Peaks 1,15(rutin),17(forsythiaside A),22(forsythin),and 26(forsythiaside)are strongly associated with the heat-clearance and detoxification effect of Green Forsythia,likely serving as the material basis for Green Forsythia’s pharmacological efficacy.
