摘 要:目的利用当归多糖(ASP)与羧甲基壳聚糖(CMCS)交联,制备可用于经皮给药系统或伤口敷料的水凝胶基质,并进行表征。方法采用柱前衍生化HPLC分析ASP的单糖组成,高碘酸钠法制备氧化当归多糖(OASP)。以L929细胞存活率、划痕愈合率为指标评价ASP氧化前后的促愈合活性。Schiff碱反应制备OASP/CMCS水凝胶,以能否成功胶凝、溶胀和保水性能、流变学性质为指标,单因素试验考察交联的影响因素,筛选最优处方,并对水凝胶的宏观性质、内部结构、溶胀性能、生物相容性以及体外生物活性等进行表征与评价。结果ASP主要由半乳糖醛酸、葡萄糖、半乳糖、阿拉伯糖组成。相较于ASP,OASP保留了促进L929细胞迁移的作用,并能促进L929细胞的增殖。最优OASP/CMCS水凝胶处方为:OASP质量分数2%、氧化度54%,CMCS质量分数5%,OASP与CMCS比例3∶7,交联pH 6.5。OASP/CMCS水凝胶呈多孔结构,具有良好的载药空间、组织黏附性、自愈合性、力学性能,在中性及碱性条件下溶胀效果更佳,且具有良好的生物可降解性和生物相容性。该水凝胶能显著促进L929细胞的增殖和迁移,具有良好的体外促愈合活性。结论本研究制备了一种具备优异性能与良好生物活性的OASP/CMCS水凝胶,为经皮给药系统及伤口敷料等医疗领域提供了一种新型安全的材料选择。Objective To prepare and characterize hydrogel matrices for percutaneous drug delivery systems or wound dressings,by cross-linking Angelica sinensis polysaccharide(ASP)with carboxymethyl chitosan(CMCS).Methods The monosaccharide composition of ASP was analyzed by PMP-HPLC.Oxidized Angelica sinensis polysaccharide(OASP)was prepared by sodium periodate method.Additionally,the survival rate and scratch healing rate of L929 cells were used as indicators to determine the changes in the healing activity of ASP before and after the oxidation.OASP/CMCS hydrogels were prepared by Schiff base reaction.Single-factor experiments were conducted to determine the optimal formulation successful gel formation,swelling properties,water retention properties,and rheological properties.The macroscopical properties,internal structure,rheological properties,dressing properties,biocompatibility and in vitro bioactivity of hydrogels were characterized and evaluated.Results ASP was composed of galacturonic acid,glucose,galactose,and arabinose.Compared with the ASP,OASP retained the ability to promote L929 cell migration and promoted the proliferation of L929 cells.The optimal OASP/CMCS hydrogel prescription was as follows:OASP mass fraction 2%,oxidation degree 54%,CMCS mass fraction 5%,OASP and CMCS ratio 3∶7,and crosslinking pH 6.5.The hydrogel exhibited a porous structure,featuring excellent drug-loading capacity,tissue adhesiveness,self-healing ability,and mechanical properties.It swelled more effectively under neutral and alkaline conditions with good biodegradability and biocompatibility.Furthermore,the hydrogel obviously promoted the proliferation and migration of L929 cells,indicating good in vitro healing activity.Conclusion OASP/CMCS hydrogel with excellent properties and biological activity is prepared,which provides a new safe choice for transdermal drug delivery system and wound dressing.
