四子定喘汤调控巨噬细胞极化改善幼龄哮喘大鼠气道炎症的实验研究

摘  要:目的探讨四子定喘汤对幼龄哮喘大鼠气道炎症及巨噬细胞极化的影响。方法将幼龄SD大鼠随机分为正常组,模型组,地塞米松组,低、中、高剂量四子定喘汤组,每组6只;除正常组外,其余各组采用卵清蛋白(OVA)激发制备幼龄大鼠哮喘模型,各组分别给予相应药物治疗。采用小动物呼吸系统检测各组大鼠呼吸频率、气道反应性(Penh值);HE染色观察大鼠肺组织病理结构变化;ELISA法检测血清白细胞介素-1β(IL-1β)、白细胞介素-10(IL-10)含量;免疫荧光检测肺巨噬细胞标志物CD86、CD163的表达;qRT-PCR、Western blot法检测大鼠肺组织巨噬细胞标志物肿瘤坏死因子-α(TNF-α)、转化生长因子-β1(TGF-β1)的水平。结果与正常组相比,模型组大鼠可见肺间质大面积充血,肺泡间隔增厚,炎性细胞浸润等病理改变,OVA激发后大鼠呼吸频率、Penh值、血清中IL-1β含量、肺组织TNF-α和TGF-β1 mRNA表达水平及CD86和CD163的表达水平明显升高(P<0.001),血清IL-10含量明显降低(P<0.001)。与模型组相比,各剂量四子定喘汤处理组和地塞米松组大鼠肺组织病理变化均有不同程度减轻,OVA激发后大鼠呼吸频率、Penh值、血清IL-1β含量、肺组织TNF-α和TGF-β1 mRNA表达水平及CD86和CD163表达水平显著下降,血清IL-10含量明显上升。结论四子定喘汤可减轻幼龄哮喘大鼠肺组织炎性细胞浸润和气道炎症,其机制可能与调控巨噬细胞极化有关。Objective To determine the effect of Sizi Dingchuan decoction on the airway inflammation and macrophage polarization in young asthmatic rats.Methods Young SD rats were randomly divided into a normal group,a model group,a dexamethasone group,and low-dose,medium-dose and high-dose Sizi Dingchuan decoction groups,with 6 rats in each group.Except for the normal group,the other groups were activated by ovalbumin(OVA)to prepare young rat asthma models,and each group was given corresponding treatment respectively.The respiratory frequency and airway responsiveness(Penh value)of rats in each group were detected by small-animal respiratory system;the pathological structure changes of rat lung tissues were observed by HE staining;the contents of serum IL-1βand IL-10 detected by ELISA;the expressions of lung macrophage markers CD86 and CD163 detected by immunofluorescence;and the levels of macrophage marker TNF-αand TGF-β1 in the rat lung tissues detected by qRT-PCR and Western blot.Results Compared with the normal group,rats in the model group showed large-area hyperemia in the lung interstitium,thickening of alveolar septa,infiltration of inflammatory cells and other pathological changes.After the OVA activation,the respiratory frequency,Penh value,the content of IL-1βin the serum,the expression levels of TNF-αand TGF-β1 mRNA in lung tissues,and the expressions of CD86 and CD163 in the rats were all significantly increased(P<0.001),while the content of serum IL-10 was significantly decreased(P<0.001).Compared with the model group,the pathological changes in the lung tissues in rats in each Sizi Dingchuan decoction treatment group and dexamethasone group showed degrees of alleviation.After the OVA activation,the respiratory frequency,Penh value,the content of IL-1βin the serum,the expressions of TNF-αand TGF-β1 mRNA in the lung tissues,and the expressions of CD86 and CD163 were greatly decreased,while the content of IL-10 in the serum was significantly increased.Conclusion Sizi Dingchuan decoction can reduce the i
