摘  要:目的建立同时测定不同产地卷柏中穗花杉双黄酮、扁柏双黄酮、selaginellin、selaginellinA、selaginellin M和selaginellin E含量的方法。方法采用超高效液相色谱-三重四极杆质谱联用(UPLC-MS/MS)法:色谱柱为WatersAcquityUPLC BEH C_(18)柱,以乙腈-0.1%甲酸水溶液为流动相梯度洗脱,流速为0.3 mL·min^(-1),柱温:30℃。采用电喷雾离子源正离子模式检测,多反应监测扫描。结果所建立的方法显示出良好的线性关系,并具备优异的精密度、重现性和稳定性。15个不同产地的卷柏在活性成分含量上存在显著差异,层次聚类分析(HCA)和主成分分析(PCA)结果显示15批样品明显分为三类。结论本研究建立了一种卷柏药材UPLC-MS/MS定量分析方法,该方法快速、准确,适用于双黄酮和炔酚类成分的测定,为其质量控制和评价提供参考依据。Objective To determine the content of amentoflavone,hinokiflavone,selaginellin,selaginellin A,selaginellin M,and selaginellin E in Selaginella tamariscina from 15 regions.Methods Ultra performance liquid chromatography-tandem quadrupole mass spectrometry(UPLC-MS/MS)method was used.The determination was performed on Waters Acquity UPLC BEH C_(18) with mobile phase consisting of acetonitrile-0.1%formic acid by gradient elution.The flow rate was 0.3 mL·min^(-1),and the column temperature was 30℃.MS detection was conducted by electrospray ionization in the positive modes.Multiple reaction monitoring mode was used for quantitative analysis.Results The method exhibited a strong linear correlation and superior precision,repeatability,and stability.Analysis of 15 samples from different origins of Selaginella tamariscina showed great variations in the content of active ingredients.Hierarchical cluster analysis and principal component analysis indicated three distinct clusters among the 15 batches.Conclusion The study successfully establishes a rapid and accurate UPLC-MS/MS quantitative analysis method for the determination of flavonoids and phenolic compounds in Selaginella tamariscina,providing a reference basis for their quality control and evaluation.
